血管紧张素转化酶2的研究进展

摘    要:2000年,科学家们发现了一种与血管紧张素转化酶(ACE)具有较高同源性的酶,命名为ACE2(或ACEH)。ACE2与ACE均具有HEXXH锌离子结合结构域,也属于肾素-血管紧张素系统(RAS),但ACE2中还具有一个特异的collectrin结构域。ACE2不仅在心血管系统中发挥了重要的功能,而且作为SARS病毒入胞的受体在肺部疾病中发挥了一定作用。此外,由于其组织特异性,可能在生殖系统及胃肠道系统中也扮演了重要的角色。

Abstract:ACE2 (or ACEH), the first homologue of human angiotensin-converting enzyme (ACE), was identified in 2000. This zinc metalloprotease belongs to the renin-angiotensin system. The ACE2 protein may evolve as a chimera of a single HEXXH zinc-binding domain and a collectrin domain. ACE2 is not only an essential regulator of heart function, but also a receptor of the SARSCoV spike glycoprotein in mediating virus entry and cell fusion. Furthermore, it may also play some roles in the physiology and pathophysiology of gastrointestinal and reproductive systems, in view of its organ-and cell-specific expression of ACE2.
