| 摘 要： | 目的初步探讨小檗碱对胃癌细胞系AGS细胞增殖和凋亡的影响。方法采用MTT的方法检测不同浓度小檗碱作用48h后对AGS细胞的抑制率，并计算半抑制浓度（50％inhibitingcon—centration，IC50）值；采用流式细胞技术检测不同浓度小檗碱作用48h后胃癌细胞系AGS凋亡的发生率和细胞周期的变化。结果小檗碱对胃癌细胞AGS生长的抑制作用呈现出剂量依赖性，48hIC。。为39．15umol／L。IC50浓度小檗碱干预48h后，AGS凋亡率发生率明显增加，与对照组比较差异有统计学意义（23．18％±3．47％vs10．05％±2．96％，P〈0．05）；IC50浓度的小檗碱作用后，AGS胃癌细胞GO／G1期细胞比例较对照组明显升高，两组比较差异具有统计学意义（P〈0．05），而S期细胞及G2／M期比例减少，但无明显统计学意义。结论小檗碱对胃癌细胞系AGS细胞具有浓度依赖性的抑制作用，抑制作用与阻滞细胞周期于G0／G1期有关，其作用机制可能与抑制DNA合成和诱导凋亡有关。
