一氧化氮合成酶在痉挛脑血管壁动态变化的实验研究

摘    要: 目的研究蛛网膜下腔出血(SAH)后痉挛的血管壁中内皮型一氧化氮合成酶(eNOS)的演变过程.方法采用兔SAH模型,全部实验动物于首次注血前和处死前行脑血管造影,取基底动脉进行免疫组织化学分析,了解eNOS的免疫表达.结果脑血管造影显示第3、5、7和10 d组注血后基底动脉直径明显减小(P<0.05),而对照组和第14 d组减小不明显(P>0.05).免疫组化结果表明,对照组见有eNOS重度染色,14 d组染色与对照组接近,第3、5、7、10 d组染色较对照组色淡,以7 d组最淡.结论SAH后痉挛期血管壁中eNOS减少,进一步证明了一氧化氮减少在SAH后脑血管痉挛(CVS)发展过程中发挥了重要作用.

Nitric oxide synthase level in rabbit basilar artery after subarachnoid hemorrhage

Abstract:Objective To investigate the levels of endothelial nitric oxide synthase(eNOS) following subarachnoid hemorrhage in rabbit basilar arteries.Methods 36 rabbits were assigned to six groups:group 1 control;group 2,sacrificed 3 days after the first intracisternal injection of blood; groups 3,4,5 and 6, given twice intra cistemal injection of blood and sacrificed 5 ,7,10, and 14 days after the first injection, respectively. Constriction of the basilar artery was confirmed by arterial angiography. Rabbit basilar arteries were analyzed by means of im-munochistochemistry for eNOS.Results In groups 2,3,4 and 5,angiography showed the basilar artery decreased in diameter, with significant differences between the two angiograms ( P < 0.05), while in groups 1 and 6, the basilar arteries decreased in diameter, but the differences were not significant between the two angiograms( P > 0.05) . On immunohistochemical study,highly positive staining was found in basilar arteries of group 1; immunohistochemical staining in group 6 was similar to that in groupl; group 2,3,4 and 5, especially group 4, showed slightly immunohistochemical staining. Conclusions Endothelial NOS decreases in rabbit basilar arteries after subarachnoid hemorrhage, the investigation indirectly proves that NO is a major pathogenic factor of cerebral vasospasm.
