Abstract:
Objective:To study the effect of BRU on apoptosis of MDA-MB-231 cells in triple negative breast cancer.Methods:MDA-MB-231 cells were treated with BRU.CCK-8 method was used to detect the growth inhibition rate of MDA-MB-231 cells.The apoptosis rate and cell cycle were detected by flow cytometry.Results:The results of the determination of growth inhibition rate of MDA-MB-231 cells in triple negative breast cancer by BRU showed that the growth inhibition rate of MDA-MB-231 cells in different concentrations of BRU was significantly different from that in the control group(all P<0.05),which was time-dependent and concentration dependent.Apoptosis results showed that the cells were treated with 5,10,20 and 40μmol/L BRU for 48 hours,the apoptosis rate of the cells increased with the increase of the concentration,and the differences were statistically significant compared with the blank control group(all P<0.05).Cell cycle results suggested that the cells were treated with 5,10,20,40μmol/L BRU for 48 h,the proportion of cells in G 0/G 1 phase gradually increased with the increase of drug concentration,and the proportion of cells in S/G 2/M phase decreased,with statistically significant differences compared with the blank control group(all P<0.05).Conclusion:Brusatol can inhibit the growth of MDA-MB-231 cells in triple negative breast cancer and induce their apoptosis in vitro.
