| 摘 要： | 应用Ｓｏｕｔｈｅｒｎｂｌｏｔ结合Ｗｅｓｔｅｒｎｂｌｏｔ分析了６１株福氏２ａ的侵袭毒力表达（ｉｐａＢＣ和ＩｐａＢＣ）。结果表明：６１株福氏２ａ的侵袭基因位于７．６ｋｂ、２．５ｋｂ和１．６ｋｂ的ＥｃｏＲＩ酶切片段。５８株广州分离株及福州分离株Ｆ００３、Ｆ４１０侵袭蛋白表达良好，而福州分离株Ｆ００８无侵袭蛋白表达，但ｉｐａＢＣ基因探针杂交阳性，显示Ｆ００８的ｉｐａ的表达调控基因有部分缺失。作者认为Ｓｏｕｔｈｅｒｎｂｌｏｔ和Ｗｅｓｔｅｒｎｂｌｏｔ分析具有特异、准确、快速的特点，不仅适用于鉴定志贺氏菌侵袭毒力表达而且可用于其它致病菌的相关毒力表达研究。 |

| Abstract: | The invasive virulence expression of 61 strains of S.flexneri 2a 30 outbreak strains and 28 sporadic strains isolated from Guangzhou in 1992. and 3 sporadic strains F003, F008 and F410 isolated from Fuzhou in 1991 .were examined by using Southern and Western blots. The ipaBC gene probe labeled with [a-32 P] dATP hybridized with three EcoR I fragments(7. 6kb . 2. 5kb and 1.6kb) of the plasmid DNAs extracted from the isolates in Southern blot. The ipa BC gene expression could be detected by Western blot easily, except for F008, in all the isolates. indicating that F008 had lost some fragments controlling the expression of ipa henes. In conclusion. the combination of southern blot and Western blot provides a simple, sensative and specific method for studing the virulence expression of S. flexneri and perhaps other bacteria. |
