补肾活髓颗粒对慢性再生障碍性贫血患者mTOR信号通路的影响

目的 :观察补肾活髓颗粒治疗慢性再生障碍性贫血(CAA)的临床疗效,并从调控m TOR信号通路角度探讨其作用机制。方法:70例患者按随机数字表法分成两组,试验组在常规西药治疗的基础上加用口服补肾活髓颗粒治疗,对照组采用西药常规治疗,并设30例健康人为正常组。观察治疗前后临床症状、临床疗效及不良反应,并用RT-PCR方法检测CAA患者骨髓单个核细胞m TOR信号通路中的m TOR m RNA表达。结果:试验组总有效率83.33%,优于对照组的64.51%(P0.05)。两组治疗后血红蛋白(HGB)、白细胞(WBC)、血小板(PLT)均有明显上升(P0.01)。CAA患者骨髓中m TOR表达水平低于正常水平(P0.01),治疗后两组m TOR m RNA表达水平均有所提高,但试验组高于对照组(P0.01)。结论:补肾活髓颗粒可显著改善CAA患者临床症状,提高临床疗效,且安全、无不良反应。补肾活髓颗粒可显著提高HGB、WBC、PLT的数量。补肾活髓颗粒通过激活CAA患者m TOR信号通路活性增加骨髓造血细胞的增殖,提高骨髓造血细胞的数量,并改善恢复骨髓造血功能。

Effect of Bushen Huosui Granules on mTOR Signaling Pathway in Patients with Chronic Aplastic Anemia

Objective:To observe the effect of Bushen Huosui granules on patients with chronic aplastic anemia(CAA),and to explore its effect on regulating the m TOR signaling pathway. Methods:There were 70 patients with CAA randomly divided into two groups. Besides routine western medicine therapy,the treatment group was also given Bushen Huosui granules. In the control group patients were given western medicine.There were 30 healthy people as the normal group. Before and after the treatment,clinical symptoms,clinical efficacy and adverse reactions were observed,and RT-PCR was applied to detect the expression of m TOR m RNA in the bone marrow mononuclear cells. Results:In the treatment group the total effective rate was83.3%,and in the control group the total effective rate was 64.51%,with statistical significance(P<0.05).And in the 2 groups after the treatment,HGB,WBC and PLT increased obviously(P<0.01). The expression level of m TOR m RNA in the patients with CAA was lower than the healthy people(P<0.01). After the treatment,in the treatment group and the control group,the expression level of m TOR m RNA increased,and in the treatment group it was higher than that in the control group(P<0.01). Conclusions:Bushen Huosui granules can significantly improve clinical symptoms in patients with CAA,improve clinical curative effect,and it is safe without adverse reaction. Bushen Huosui granules can significant improve the levels of HGB,WBC and PLT. Bushen Huosui granules increases the proliferation of bone marrow hematopoietic cells by activating m TOR signaling pathway to increase the number of bone marrow hematopoietic cells,and to promote the recovery of bone marrow hematopoietic function.
