督脉灸疗法对阿尔茨海默病模型小鼠学习记忆能力及海马CA1、CA3区HE染色的影响

摘    要:目的:探讨督脉灸疗法(督灸)对阿尔兹海默病模型小鼠学习记忆能力及海马CA1、CA3区HE染色的影响。方法:将40只SAMP8小鼠随机分为正常组、模型组、督灸组、中药组和督灸+中药组各8只,其中督灸组采用督灸疗法,中药组给予六味地黄丸灌胃,实验结束后,采用Morris水迷宫实验法进行行为学检测,HE染色法检测CA1、CA3区组织形态学改变。结果:与模型组相比,中药组、督灸组、督灸+中药组的小鼠逃避潜伏期均降低(P0.05或P0.01),其中督灸组和督灸+中药组用时最短,与中药组比较(P0.05或P0.01),表明督灸疗法可提高SAMP8小鼠学习记忆能力,且疗效优于中药组;HE染色结果显示,相比正常组,模型小鼠CA1、CA3区细胞组织均有坏死损伤,相比模型组,督灸组和督灸+中药组CA1、CA3区细胞坏死数量最少,形态学改变最轻,中药组损伤较为明显,说明督灸疗法对SAMP8小鼠海马CA1、CA3区神经元具有保护作用,可减少细胞损伤。结论:督灸疗法能提高SAMP8小鼠学习记忆能力,其机制与督灸能保护海马CA1、CA3区神经元和抑制细胞损伤有关。

Effect of Governor-Moxibustion on Learning and Memoryand HE Staining in Hippocampal CA1 and CA3 Area of Mouse with Alzheimer's Disease

Abstract:Objective:To investigate the effect of governor-moxibustionon learning and memory ability and HE staining in hippocampal CA1 and CA3 of Alzheimer's disease model mice. Methods:Forty SAMP8 mice were randomly divided into normal group,model group,governor-moxibustion group,Chinese herbal medicinegroup and governor moxibustion plus Chinese herbal medicine group,8 mice in each group. In the governor-moxibustion group the mice were given governor-moxibustion and in the Chinese herbal medicine group the mice were given Liuwei Dihuang pill through gavage. At the end of the experiment,Morris water maze test was used to conduct behavioral testing. HE staining was used to detect the histological changes in CA1 and CA3 areas. Results:Compared with the model group,the escape latency of the Chinese herbal medicine group,governor-moxibustion group and governor moxibustion plus Chinese herbal medicine group were all decreased(P <0.05 or P <0.01),and that of governor-moxibustion group and the governor-moxibustion plus Chinese herbal medicine group were the shortest,indicating that governor-moxibustioncan improve learning and memory ability of SAMP8 mice,and the curative effect is better than that of Chinese herbal medicine group(P<0.05 or P <0.01);the results of H.E. staining showed that compared with the normal group,the necrosis of the cells in the CA1 and CA3 regions was observed in the model mice;compared with the model group,the number of cell necrosis in the CA1 and CA3 areas in the governor-moxibustion plus Chinese herbal medicine group was the least,with the lightest morphological changes,while in the Chinese herbal medicine group,the injury was more obvious,indicating that the governor-moxibustion can protect hippocampal CA1 and CA3 neurons in SAMP8 mice,and can reduce cell damage. Conclusion:Governor-moxibustioncan improve the learning and memory ability of SAMP8 mice. The mechanism may be related to governor-moxibustion protecting neurons in hippocampal CA1 and CA3 area and inhibiting cell damage.
