隔药脐疗对卵巢早衰大鼠VEGF和AMH表达的影响

目的:探讨隔药脐疗法对卵巢早衰(POF)大鼠血管内皮生长因子(VEGF)及抗苗勒氏管激素(AMH)的作用机制。方法:50只Wistar雌性大鼠随机分成正常组、隔药脐疗组、模型组、西药组,采用腹腔注射环磷酰胺制作大鼠卵巢早衰模型。隔药脐疗组采用自制灸膏做脐疗,西药组采用补佳乐灌胃。治疗2周后,采用放射性免疫法检测大鼠血清雌二醇(E2)、促黄体生成素(LH)及促卵泡激素(FSH)水平,应用酶联免疫吸附法检测血清VEGF及AMH的含量。结果:与正常组相比,模型组E2水平降低(P<0.05),与模型组比较,隔药脐疗组和西药组均升高(P<0.05)。与正常组比较,模型组大鼠血清中VEGF及AMH含量明显下降(P<0.05);与模型组比较,隔药脐疗组及西药组血清VEGF及AMH水平显著升高(P<0.05);隔药脐疗组与西药组之间VEGF和AMH水平差异无统计学意义(P>0.05)。结论:隔药脐疗法对卵巢早衰大鼠的卵泡数量及卵巢储备功能均有明显的促进作用,可能与其对血清E2、VEGF和AMH的表达调节有关。

Influence of Herb-Partition Moxibustion at Navel on Expression of VEGF and AMH in Rats with Premature Ovarian Failure

Objective:To investigate the mechanism of herb-partition moxibustion at navel on vascular endothelial growth factor(VEGF) and anti-mullerian hormone(AMH) in rats with premature ovarian failure(POF). Methods:Fifty female Wistar rats were randomly divided into normal group,model group,herb-partition moxibustion at navel group and western medicine group. The rat model of POF was established by intraperitoneal injection of cyclophosphamide. The rats in the herb-partition moxibustion at navel group were treated with self-made herb ointment. The rats in the western medicine group were given estradiol valerate by gavage. After 2 weeks of treatment,the levels of E2,LH and FSH were detected by using the radioimmunoassay method. Enzyme-linked immunosorbent assay(ELISA) was used to detect the level of VEGF and AMH. Results:Compared with the normal group,the level of E2 decreased in the model group(P<0.05). The level of E2 in the herb-partition moxibustion at navel group and western medicine group was higher than that in the model group(P<0.05). Compared with the normal group,the level of VEGF and AMH significantly decreased in the model group(P<0.05). The level of VEGF and AMH in the herb-partition moxibustion at navel group and western medicine group increased,compared with the model group(P<0.05). There was no statistical difference of the level of VEGF and AMH between the herb-partition moxibustion at navel group and western medicine group(P> 0.05). Conclusion:The herb-partition moxibustion at navel can significantly promote the follicle number and ovarian reserve of POF rats,which may be related to the its regulation on E2,VEGF and AMH.
