加味膈下逐瘀汤对肝纤维化模型大鼠肝功能及纤维化指标的影响

摘    要:目的:研究加味膈下逐瘀汤抗肝纤维化的效果及作用机制。方法:将大鼠分为正常组、模型组、秋水仙碱组及加味膈下逐瘀汤组,每组10只。秋水仙碱组按1 mg/kg药量给予秋水仙碱,加味膈下逐瘀汤组按生药量8.2 g/kg灌胃,正常组及模型组分别给予相同体积生理盐水。考察各组肝脏指数、血清肝功能和肝纤维化指标,并进行HE染色,观察肝组织病理改变。结果:与模型组比较,加味膈下逐瘀汤组大鼠肝纤维化明显改善,肝脏指数降低,丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、层粘连蛋白(LN)和透明质酸(HA)水平降低,清蛋白(ALB)和总蛋白(TP)含量升高,差异有统计学意义(P0.05);组织病理学结果显示,加味膈下逐瘀汤能明显改善肝脏病理损伤程度。结论:加味膈下逐瘀汤能有效减轻肝纤维模型大鼠的肝脏损伤,降低肝纤维化程度。

Effect of Jiawei Gexia Zhuyu Decoction on Liver Function and Hepatic Fibrosis Index of Model Rats of Hepatic Fibrosis

Abstract:Objective:To study the effect of Jiawei Gexia Zhuyu decoction(JGZD) on liver fibrosis and its action mechanism. Methods:The rats were divided into normal group,model group,colchicine group and JGZD group,with 10 rats in each group. The colchicine group was given colchicines and the dosage was 1 mg/kg.The JGZD group was given JGZD by gavage and the dosage was equal to that of crude medicine. The normal group and the model group were given saline of the same volume. The organ coefficients of liver was tested and the serum was inspected for getting liver function and liver fibrosis index. The sections of liver tissues were stained by HE to observe the histopathological changes. Results:Compared with the model group,the liver fibrosis of rats in the JGZD group were significantly improved;the organ coefficients of liver of rats in JGZD group were significantly decreased;the levels of alamine aminotransferase(ALT),aspartate aminotransferase(AST),laminin(LN) and hyaluronic acid(HA) in serum of rats in the JGZD group were significantly decreased;the levels of albumin(ALB) and total protein(TP) in serum of rats in the JGZD group were significantly increased. All the above differences were statistically significant(P<0.05). The histopathological results showed that JGZD could obviously alleviate the pathological damage of the liver. Conclusion:JGZD can alleviate the damage of the liver and reduce the degree of fibrosis of model rats.
