| 摘 要： | 目的:观察自拟中药复方"益肾蠲湿合剂"对肾小球系膜细胞(glomerular mesangial cell,GMC)增殖的影响,并进一步探讨其作用机理。方法:将1×10-3、1×10-2和1×10-1mg/mL的"益肾蠲湿合剂"提取液作用于体积分数10%血清的DMEM培养的GMC,分别用MTT和Western Blot方法检测细胞的增殖和磷酸化ERK1/2的表达。结果:1×10-2和1×10-1mg/mL"益肾蠲湿合剂"可显著抑制GMC的增殖,抑制率在12、24和36 h分别为13.4%、为9.6%。细胞总ERK的表达在各组间无明显差别。但不同浓度的"益肾蠲湿合剂"能显著地抑制血清所诱导的ERK1/2的磷酸化,最高抑制率达68%。结论:"益肾蠲湿合剂"可通过抑制ERK1/2的磷酸化来抑制GMC的增殖。 |
