生长分化因子-9在卵巢反应不良患者颗粒细胞中的表达

摘    要:
目的探讨生长分化因子-9(GDF-9)在卵巢反应不良患者颗粒细胞的表达及其在超促排卵中卵泡发育的作用。方法对49例接受体外受精-胚胎移植/卵胞浆内单精子注射(IVF-ET/ICSI)治疗的患者,给予相同的改良短方案超促排卵,采用免疫组织化学方法测定颗粒细胞中GDF-9水平,比较卵巢反应不良组(实验组,n=30)与正常对照组(对照组,n=19)GDF-9表达的差异。结果两组颗粒细胞中均测及GDF-9的表达,实验组颗粒细胞中GDF-9的表达显著低于对照组(P0.05)。结论颗粒细胞中GDF-9对控制性超排卵中卵泡的生长发育可能具有调节作用,且卵巢反应不良患者颗粒细胞中GDF-9表达降低,可能参与卵巢反应不良的发病机制。

Abstract:
Objective: To study the expression of growth differentiation factor-9 (GDF-9) in human granulosa cells of poor ovarian responders in controlled ovarian hyperstimulation (COH). Methods: Forty-nine samples of human granulosa cells were obtained from patients undergoing IVF- ET/ICSI treatment. All of the patients were stimulated with improved GnRH-a short protocols. GDF-9 expression in human granulosa cells was determined by immunohistochemical method. 49 samples were divided into two groups according to the ovaian response to the stimulation: poor responder group (n= 30) and normal responder group (n=19). Results: GDF-9 was detected in all human granulosa cell samples. The expression of GDF-9 was significantly higher in normal responder group than that in poor responder group (P~0.05). Conclusions: It is suggested that the expression of GDF-9 in human granulosa cells may play a crucial role in follicle development, and the decreased expression may be related to the poor ovarian response to COH.
