局部植入接骨中药提取物促进骨愈合的实验研究

摘    要:目的 评价局部植入接骨中药对颅骨缺损愈合的影响。方法 雄性Wistar大鼠32只,随机分两组,每组16只。应用接骨中药提取物与经盐酸胍处理的无骨诱导活性的不溶性骨质(Insoluble bone matrix,IBM)为植入材料,将中药提取物IBM复合植入直径8lmm的大鼠颅骨缺损和胸部皮下组织内。不含中药提取物的IBM植入上述部位(不同动物)作为对照。术后10d和30d取植入物行组织学、组织化学和生物化学分析。结果 术后10d,植入中药提取物IBM的颅骨缺损内见间充质细胞向成骨细胞分化的形态特征。术后30d,中药提取物:IBM复合物在大鼠颅骨缺损内形成大量的新骨,且大多位于骨缺损中部,与宿主骨不相连接;单纯植入IBM的颅骨缺损中部未见新骨形成。中药提取物-IBM和单纯IBM在胸部皮下组织内未能诱导异位成骨。结论 接骨中药提取物植入颅骨缺损部位后能促进间充质细胞向成骨细胞分化,从而加速骨缺损的愈合。

Abstract:Objective To evaluate the effects of local implantation of bone-healing herb extract on the healing of cranial bone defects Methods32 male Wistar rats were randomly divided into two groups,16 rats in each group.Extract of TCM and no-inductive insoluble bone matrix(IBM)after guanidine-HCI extraction regard as implanted material,which were embed in skull defects and subcutaneous tissue of chest of rats.Alone IBM was embeded in the same site as control group(in different rats).Histology,histochemistry and biochemistry of the implants were analyzed at 10th and 30th days after implantation.Results After 10 days,mesenchymal cells to osteoblasts disserentiation with herb extract-IBM complex preparation were observed.After 30 days,abundant new bone formation occurred in the skull defects with herb extract-IBM complex preparation.Above-condition was not found in control group.Conclusion Local implantation of bone-healing herb extract into skull defects can enhance differentiation of mesenchymal cells to osteoblasts and accelerates the healing process of bone defects.
