骨髓间充质干细胞移植治疗脊髓损伤

摘    要:目的:探讨大鼠骨髓间充质干细胞(MSCs)对脊髓损伤与修复的作用与机制及细胞移植的方法与途径。方法:实验组为将体外培养SD大鼠的MSCs经绿色荧光蛋白(GFP)转染标记后显微注射于脊髓横断模型的脊髓损伤处,并以无血清培养基注射动物为对照组。12周后观察移植细胞的分化情况,并通过行为学、组织学及免疫组化的方法评价脊髓功能变化。结果:实验组MSCs移植12周后,脊髓功能明显恢复。结论:骨髓间充质干细胞移植对脊髓的损伤修复具有明显作用。

Abstract:Objective To investigate the methods that how the MSCs wok on the spinal cord injury. And try to find the ways to plant MSCs. Methods The MSCs were transplanted into the spinal cord of SCI modles. Before the transplanting, the MSCs were transfected with EGFP.12 weeks after transplantation, MSCs' difference were observed, SPECT, immunohistochemistry and angiogenesis were observed to detect the effects on spinal cord. Results This experiment obtained the high pure MSCs. The MSCs can stabilize to spread the generation to combine the cord. Conclusion The transplantation using the cells proved that MSCs was very useful in the recovery of spinal cord injury Which may allow us to rebuild those conditions to repair and regrow the spinal cord.
