中医三种治法对人肝癌细胞增殖及端粒酶活性的影响

摘    要:目的:探讨中医活血软坚、益气养阴、清热解毒三种治法影响肝癌细胞增殖及端粒酶活性的特点和意义.方法:从临床治疗肝癌常用的活血软坚药中选取有代表性的丹参、川芎、鳖甲,益气养阴药中选取黄芪、枸杞子、女贞子,清热解毒药中选取半枝莲、白花蛇舌草,分别制成水煎剂,灌喂正常大鼠,采血制备药物血清.以正常鼠血清为阴性对照,将药物血清温育Bel-7402人肝癌细胞.噻唑蓝比色法测定细胞增殖,多聚酶链反应酶联免疫吸附(PCR-ELISA)法检测细胞端粒酶活性.结果:活血软坚药和益气养阴药均可抑制Bel-7402细胞的增殖,清热解毒药不能抑制Bel-7402细胞的增殖.活血软坚药、益气养阴药和清热解毒药均可抑制Bel-7402细胞端粒酶活性,且活血软坚药作用优于清热解毒药.结论:活血软坚法和益气养阴法均可抑制Bel-7402细胞的增殖,其主要机制可能是通过抑制Bel-7402细胞端粒酶活性.清热解毒法不能抑制Bel-7402细胞的增殖,但有一定的抑制细胞端粒酶活性的作用.

Abstract:To investigate the influence o three Chinese medical therapies (activating blood and dissipating a mass, benefiting qi and nou(?)ishing yin, clearing away heat and toxic materials) on the proliferation and telomerase activity of human hepatocarcinoma cells. Methods: From the clinical frequently applied herbs for hepatoca(?)cinoma, Radix salvia miltiorrhiza, Rhizoma chuanxiong, Carapax trrionycis were chosen as the typical drugs of activating blood and dissipating a mass, Radix as tragali, Fructus lycii, Fructus ligustrum lucidi as the typical drugs of benefiting qi and nourishing yin, Herba scutellariae barbatae, Herba hedyotis diffusae as the typical drugs of clearing away heat and toxic materials. These drugs were prepared as decoction respectively, and fed to the normal rats. The drug sera were collected from the rats and incubated with cultured Bel-7402 human hepatocarcinoma cells. While the normal rat sera were used as the control group. The proliferation of Bel-7402 cells was observed by MTT colorimetric assay, the telomerase activity of Bel-7402 cells was tested by PCR-ELISA method. Results: Both the groups of activating blood and dissipating a mass and benefiting qi and nourishing yin inhibited the proliforation of Bel-7402 cells. The group of clearing away heat and toxic materials didnot inhibit the proliferation of Bel-7402 cells. All of the three groups suppressed the telomerase activity of Bel-7402 cells, and the effect of group of activating blood and dissipating a mass was better than that of group of clearing away heat and toxic materials. Conclusion: Both the therapies of activating blood and dissipating a mass and benefiting qi and nourishing yin can inhibit the proliferation of Bel-7402 cells, the main mechanism of which may be suppressing the telomerase activity of Bel-7402 cells. The therapy of clearing away heat and toxic materials can not inhibit the proliferation of Bel-7402 cells, but can suppress the telomerase activity of Be1-7402 cells in some degree.
