HPLC法测定消积丸中大黄素和大黄酚的含量

目的：建立消积丸中大黄素与大黄酚的含量测定方法。方法采用Thermo Syncronis C18色谱柱（4.6mm ×250mm，5μm），以甲醇-0.1％磷酸（85∶15）为流动相，流速：1.0mL· min －1，柱温：30℃，检测波长：254nm。结果大黄素和大黄酚进样量分别在0.0056～0.2236μg（r ＝0.9999）和0.0155～0.6207μg（r＝0.9999）范围内与各自峰面积积分值呈良好的线性关系；精密度、稳定性、重复性试验的 RSD均≤1.51％；平均加样回收率分别为99.49％、99.68％，RSD分别为0.89％、0.38％（n＝6）。结论该方法简便、快捷、准确，重现性好，可作为消积丸的质量控制方法。

Detemination of the Contents of Emodin and Chrysophanol in Xiaoji Pills by HPLC

ABSTRACT:OBJECTIVE To establish an HPLC method for the determination the contents of emodin and chry-sophanol in Xiaoji Pills.METHODS The separation was carried out on a Thermo Syncronis C18 column (4.6mm × 250mm,5μm) with methanol-0.1%phosphoric acid solution (85∶15).The flow rate was 1.0mL· min-1.The de-tection wavelength was 254nm.The column temperature was 30℃.RESULTS The linear ranges of emodin and chrysophanol were 0.0056~0.2236μg(r=0.9999) and 0.0155~0.6207μg(r=0.9999).The average recoveries were 99.58%and 99.68%with RSDs of 0.89%and 0.38%(n=6).RSDs of precision,stability and reproducibili-ty tests were lower than 1.51%.CONCLUSION The method is simple,quick,accurate and reproducible,and can be used for the qulity control for Xiaoji Pills.
