摘    要:目的 探讨促红细胞生成素 (erythropoietin ,EPO)进行预处理对缺血海马CA1区神经元是否具有保护作用。方法 采用 4 -VO法制作全脑缺血模型 ,将SD大鼠分为假手术组、生理盐水对照组、EPO处理组。生理盐水对照组和EPO处理组于术前 3h ,分别脑室立体定向注射生理盐水和重组人促红细胞生成素 (rHu -EPO) ,假手术组只进行假手术处理。观察缺血后 72h大鼠的生存情况及海马细胞凋亡和超微结构。结果 EPO处理组海马CA1区较生理盐水对照组有较少的凋亡神经元 (P <0 0 1)。结论 EPO预处理可以对缺血后海马CA1区神经元产生保护作用。

Abstract:Objective To investigate whether pretreatment of erythropoietin(EPO) exerts protection on neuron in hippocampal CA1 subregion. Methods Transient global brain ischemia model was established using 4-VO technique. Male Sprague Dawley rats were randomized into sham operation group, normal saline group and erythropoietin group. At 3 hours before global cerebral ischemia, saline or EPO was injected into the lateral ventricle of rat, with the assistance of stereotaxic coordinates, upon designed conditions. The Sham rats only had incision on cervix without any treatment of the arteries. Survival of rats, neuron apoptosis and electronic microphotograph were observed 72 hours after global brain ischemia in hippocamp CA1 subregion.Results There were less apoptotic neurons in EPO group than in physiological saline group(P<0.01). Conclusion Pretreatment of EPO can protect neuron against ischemia in hippocamp CA1 subregion in rat.
