参附注射液对缺氧缺血性脑损伤新生大鼠半胱氨酸蛋白酶-1的影响

摘    要:目的:探讨参附注射液(SFI)对缺氧缺血性脑损伤(HIBD)新生大鼠脑组织中半胱氨酸蛋白酶-1(Caspase-1)蛋白和mRNA的表达。方法:制备新生大鼠HIBD模型,将新生7日龄Sprague-Dawley(SD)大鼠随机分为假手术组(S)、生理盐水对照组(C)、参附组(SF),每组按术后观察时间点不同进一步分为3 h、12 h、24 h、6天、14天5个亚组,采用RT-PCR方法检测病变侧大脑皮层Caspase-1 mRNA的表达,用免疫组织化学法行Caspase-1免疫阳性细胞计数。结果:C组和SF组Caspase-1 mRNA和Caspase-1免疫阳性细胞计数在24 h、6天、14天均较S组升高(P<0.01)。24 h开始增加,6天达到高峰,随后开始下降,但14天仍高于S组(P<0.01);SF组Caspase-1 mRNA和免疫阳性细胞计数在24 h、6天、14天较C组减少(P<0.01)。结论:参附注射液(SFI)下调HIBD新生大鼠Caspase-1蛋白和mRNA的表达水平,SFI对新生大鼠HIBD有保护作用。

Abstract:Objective:To investigate the effect of Shenfu injection(SFI) on the expression of Caspase-1 mRNA and protein in neonatal rats with hypoxic-ischemic brain damage(HIBD),and clarify the protection of SFI for hypoxic-ischemic(HI) brain damage from molecule level and gene level.Methods:Seven-day-old Sprague-Dawley(SD) rats were randomly assigned to sham operation group(S),normal sodium control group(C) and Shenfu injection treatment group(SF),and Models of postnatal SD rats with HIBD were established with Rice method.They were further divided into five subgroups(16 rats per subgroup) according to the time points 3h,12h,24h,6 d and 14 d.In each subgroup,8 rats were used to measure caspase-1 mRNA in the cerebral cortex by semi-quantitative RT-PCR;another 8 rats were used to study the protein expression for caspase-1 by immunohistochemistry.Results:The expression of caspase-1 mRNA and protein in ischemic cortex increased progressively in group C and group SF from 24 h to 6 d(P<0.01 vs S),began to increase at 24 h after HI insult(P<0.01 vs.S),peaked at 6 d after HI insult(P<0.01 vs.other groups) and decreased after this.But the mRNA/protein for caspase-1 in group C and group SF was still higher than group S(P<0.01 vs.S) at 14 d,the mRNA/protein for caspase-1 after HI insult in group SF was lower than group C at same time point after 12h(P<0.01 vs.C).Conclusion:The expression of caspase-1 mRNA and protein has increased after HI insult in neonatal rats and SFI can decline the expression of caspase-1 mRNA and protein.SFI has protective function of neonatal rats with HIBD.
