BDNF及受体TrkB在大鼠小脑的表达研究

目的:研究BDNF及其受体TrkB在大鼠小脑时空分布变化。方法:进行免疫组织化学实验,观察BDNF及TrkB在正常大鼠发育期、成年期和老龄期小脑内的定位分布及时间变化。结果:发育期小脑皮质的外颗粒层、梨状细胞层和内颗粒层均有BDNF表达,梨状细胞层的阳性Purkinje细胞染色明显。成年期内颗粒层阳性细胞逐渐增多,梨状细胞层的Purkinje细胞BDNF表达及阳性细胞数量达高峰。老年期阳性细胞染色强度呈增龄减低,Purkinje细胞发生了轻度的退行性变,TrkB表达在小脑皮质分布模式类似BDNF的表达。结论:BDNF及TrkB表达的时空变化始终与小脑皮质的发生发育过程相关。生后0天,二者在小脑皮质外颗粒层、梨状细胞层和内颗粒层均有较强表达,以梨状细胞层为主。第10天左右,小脑皮质发育为典型的三层结构,Purkinje细胞表达BDNF及TrkB最为显著。成年后,表达水平保持较稳定。老龄后明显减弱,BDNF及TrkB阳性Purkinje细胞发生轻度的退行性改变。

Study the Expression of Neuiotrophin Factor and Tyrosine Kinase Receptor in Rat Cerebellum

Objective Study systematically the distribute of BDNF and TrkB in the normal rat cerebellum.Methods Observe systematically the expression changs of BDNF and TrkB in the normal rat brains during devolopement phase and adulthood and aged phase and thire relationship.Results BDNF and TrkB were expressed in cerebellum during devolopement phase The external granule layer became molecular layer along with the migration of granule cells to the internal granule cell layer,and the immuostaining cells increased gradully during adulthood.phase The Purkinje immuoreactive cells appear to peak.BDNF and TrkB were expressed significantly in Purkinje cells during adulthood.During aged phase,Purkinje cells slightly degenerated.Conclusion The expression of BDNF and TrkB reached a peak in cerebellum of rats on PND 20.The expression of BDNF and TrkB decreased gradually after adulthood.During aged phase,the expression of BDNF and TrkB increased significantly,and the form of the cell celloccured degeneration.
