依普利酮对自发性高血压大鼠心肌缝隙连接蛋白Cx43表达的影响及干预

摘    要:
目的观察自发性高血压大鼠心肌缝隙连接蛋白(connexin,Cx43)含量和分布的变化以及依普利酮对其的干预机制。方法取20只自发性高血压(SHR)大鼠(8周雄性、体质量200g)分为高血压(A)组、依普利酮(B)组,再取同周龄、同体质量普通Wistar大鼠(C)。分别于喂养2、4、6、8周测大鼠尾动脉压,喂养8周后处死,取出心脏,用免疫组织化学方法显示大鼠心肌Cx43的分布特征及Cx43含量的变化。结果与C组相比,A组大鼠心房肌及心室肌Cx43含量均较C组显著减少(分别为P<0.05和P<0.01),且分布紊乱;而B组Cx43含量较A组显著增多(P<0.01),且分布不规律程度减轻。结论醛固酮水平的升高可能导致心肌Cx43重新分布及表达减少;依普利酮能有效减轻Cx43的表达减少及重构。

Abstract:
Objective To investigate the effect of eplerenone and content and distribution changes spontaneity hypertension rats myocardium gap junction of connexin 43(Cx43).Methods Twenty male SHR with eight weeks old and two hundred grams weight were divided into hypertension group(A),eplerenone group(B) and Wistar rats(with the same age and weight) group(C).To measure the rats arterial pressure of tail after feeding two,four,six and eight weeks respectively.After eight weeks.To measure the inside diameter and the mass ration of left venticle.Cx43 was detected with immunohisto-chamical method and was semi-quantitatively analyzed in different group.And then analyzed the changes of Cx43.Results The Cx43 protein level of atrial and ventricular tissue in group.A decreased significantly(P0.05 and P0.01,respectively) with inisdrstribution compared with those in group C.Cx43 level increased significantly(P0.01) in group B compared with that in group A and the inhomogeneity in Cx43 distribution decreased.Conclusion The results show that Cx43 remodeling with redistribution and reduction of protein expression may be induced by the increase of aldosterone concentration and eplerenone attenuates Cx43 expression and remodeling effectively.
