银杏酮酯对大鼠离体心肌缺血再灌注损伤的影响

摘    要:目的 观察银杏酮酯对大鼠离体心肌缺血再灌注损伤的影响,并探讨其保护机制.方法将32只SD大鼠随机分为银杏酮酯3个剂量(12.5、25、50 mg/L)组和对照组.用Krebs-Henseleit(K-H)液进行离体心脏Langendorff灌流,各组均平衡灌注30 min,停灌造成全心缺血30 min,再恢复空白K-H液(或加药K-H液)灌流40 min.再灌注期间测定血流动力学指标及心肌超氧化物歧化酶(SOD)、乳酸脱氢酶(LDH)含量.结果 用银杏酮酯灌流缺血的心脏后,与对照组比较,左室收缩压、左室功能、左心室内压最大变化率、冠脉流量的百分恢复率分别增加;冠脉流出液中SOD含量百分率明显增加,而LDH含量百分率明显减少.结论 银杏酮酯对离体的缺血再灌注损伤心脏有保护作用,其机制可能与抗氧自由基损伤有关.

Abstract:Objective To discuss the mechanism of cardioprotective effect of Ginkgo bitoba extract 50(GBE50) on ischemia-reperfusion injury in isolated rat hearts.Methods Thirty-two rats were randomly divided into three dosage of GBE50 groups(12.5,25,50 mg/L) and control group.After the heart was given Langendorff perfusion with Krebs-Henseleit(K-H) juice,the perfusion was stopped for 30 min to induce global ischemia,then it was reperfused with K-H juice or K-H juice with drug for 40 min.By using a modified Langendorff model with modified Krebs-Henseleit buffer at 37 ℃,the hemodynamic parameters,myocardial SOD and LDH were monitered.Results After GBE50 was given in reperfusion period,in isolated perfused rat heart models,LVSP,left ventricular function and ±dp/dt max of the heart increased.The average volume of coronary efflux increased respectively compared with that of control group of ischemia-reperfusion.The activities of LDH in effluent samples of GBE50 group was lower and SOD was higher than control group.Conclusion GBE50 has myocardial protective effects on ischemia-reperfusion injury in isolated rat hearts.The mechanisms of which may be associated with deactivating oxygen free radicles.
