全反式维甲酸诱导分化脑胶质瘤干细胞治疗胶质瘤的实验研究

摘    要:
目的:探讨全反式维甲酸对大鼠C6脑胶质瘤细胞增殖的抑制作用及其机制。方法:建立大鼠C6脑胶质瘤模型,腹腔注射全反式维甲酸后,采用MRI检测胶质瘤的体积变化、流式细胞仪检测细胞增殖时相变化及免疫组化检测p27Kipl蛋白的变化。结果:全反式维甲酸组较对照组肿瘤体积明显减小(P<0.05);Go期细胞比例明显增加(P<0.05),S期细胞比例明显减少(P<0.05);P27KiPl蛋白的表达明显增加(P<0.05)。结论:全反式维甲酸通过上调p27Kipl蛋白的表达而抑制大鼠C6脑胶质瘤细胞增殖。

Experimental study of differenti therapy by all-trans retinotic acid on glioma stem cells for glioma

Abstract:
Objective To study the cell prolifation inhibition and mechanism of all-trans retinotic acid on C6cells of rats. Methods The model of C6glioma cells on rats was made. After intraperitoneal injection of all-trans retinotic acid,the change of volume was detected by MRI,the phase change of cell proliferation was detected by FCM,the change of p27Kipl was detected by IHC. Results In all-trans retinotic acid group,the tumor volume reduced significantly( P〈0. 05),the ratio of Go-phase cell increased significantly( P〈0. 05),the ratio of S-phase cell reduced significantly( P〈0. 05),the expression of p27Kipl increased significantly( P〈0. 05). Conclusion All-trans retinotic acid inhibits the cell multiplication of C6cells by increasing the expression of p27Kipl.
