摘 要: | 目的建立西藏不同产区27批冬虫夏草水提液的高效液相色谱指纹图谱,并定量测定其6种主要核苷类成分(尿苷、肌苷、鸟苷、胸苷、腺苷、2'-脱氧腺苷)的质量分数。方法采用高效液相色谱法,色谱柱为Agilent Eclipse XDB—C18柱(250mm×4.6mm,5μm),检测波长260nm,以甲醇.水为流动相进行梯度洗脱,柱温25℃,流速O.8mL·min^-1。结果建立了冬虫夏草水提液的HPLC指纹图谱,包含11个共有峰,各峰分离良好,指认出尿苷、肌苷、鸟苷、胸苷、腺苷、2’-脱氧腺苷6个特征成分.同时测定了其质量分数;6种特征成分质量浓度分别在10.71-96.4(r=0.9999),8.50~76.41(r:0.9999)。7.838-70.54(r=0.9999),1.120-10.08(r=0.9998),5.715-51.44(1-=0.9999),0.2625~2.363μg·mL^-1(r=0.9997)范围内与峰面积呈良好的线性关系;平均回收率依次为96.5%、97.7%、97.5%、95.2%、96.8%、101.5%,RSD分别为2.3%、2.7%、2.0%、2.6%、3.0%、2.7%(n=6)。结论本试验建立的指纹图谱可同时对冬虫夏草的6种主要核苷类成分进行分析,对评价藏产冬虫夏草的质量具有重要意义。

Abstract: | Objective To establishan HPLC fingerprint of aqueous extract of Ophiocordyceps sinensis from the main producing area of Tibet, and explore a method to determine 6 nucleosides components ( uridine, inosine, guanosine, thymidine, adenosine and 2'-deoxyadenosine). Methods The HPLC analysis was performed on Agilent Eclipse XDB-C18 column (250 mm × 4.6 mm,5 μm) and detected at 260 nm. The mobile phase was consisted of methanol and water using a gradient elution. The column temperature was 25 ℃ and the flow rate was 0.8 mL · min^-1. Results The HPLC fingerprint was established, and eleven common characteristic peaks had a satisfactory peak separation. 6 components, including uridine, inosine, guanosine,thymidine, adenosine and 2'-deoxyadenosine, were identified and determined. The linearity of the 6 components were in 10.71-96.4(r= 0.999 9) ,8.50-76.41 (r=0.999 9) ,7.838-70.54(r=0. 999 9), 1.120-10.08(r= 0.999 8),5.715-51.44(r=0.999 9),0.262 5-2.363 μg · mL^-1(r= 0.999 7), respectively. The average recovery were 96. 5%, 97.7%, 97.5%, 95.2%, 96.8% and 101.5%, and the RSD were 2. 3%, 2. 7%, 2.0%, 2. 6%, 3.0% and 2.7% ( n = 6). respectively. Conclusion The HPLC fingerprint was established, and it provides a method for the simultaneous analysis of 6 main nucleosides components in 0. sinensis,which is important for the quality evaluation of 0. sinensis from Tibet. |
