单克隆抗体治疗多发性骨髓瘤的研究

目的研究抗多聚体蛋白聚糖单克隆抗体对人多发性骨髓瘤的治疗作用。方法选取本地区医院住院的多发性骨髓瘤（MM）患者，筛选出人多发性骨髓瘤MM细胞株YD-α和YD-β，所有患者均符合第三版张之南、沈悌主编的《血液病诊断及疗效标准》，单克隆抗体免疫组化法测定骨髓瘤细胞特异性表型，于第1天、第2天、第3天使用台盼蓝染色，并进行活细胞记数。结果间接免疫荧光标记检测分析显示，单克隆抗体A2b对YD-α和YD—β细胞表达的多聚糖蛋白分子具有较强识别作用，且A3b与YD-α和YD-β细胞膜上荧光标记检测阳性率具有相似性，A2b对YD-α和YD—β细胞生长抑制随浓度增加和随时间延长加强，P〈0．05具有显著性差异。结论单克隆抗体对MM细胞株生长有抑制作用，因此单克隆抗体疗法可能成为治疗多发性骨髓瘤的-种新手段。

Research on multiple myeloma treatment by Monoclonal antibodies

Objective To research the treatment of Monoclonal antibodies on Multiple myeloma patients. Methods Analyzed Polymer proteoglycans expression on YD-α and YD-β cell membrane by indirect immunofluoreseent label method and flow cytometry method, recorded the number of the living cells to observe the growth of on YD-α and YD-β cells. Results Polymer proteoglycans were expressed on YD-α and YD-β cells, the growth of YD-ct and YD-β ceils were strongly inhibited by A2b. Conclusions Inhibition of MM ceils by A2b may provide a new method for MM biotherapy.
