缺氧对小鼠心肌琥珀酸脱氢酶、Ca~(2 )-Mg~(2 )-腺苷三磷酸酶活性及超微结构的影响

摘    要:
采用雌雄兼有的成年小鼠40只,分4组分别置入合常氧、12%、8%、4%氧的密闭容器内1O~15min.经心脏取血测动脉血氧分压,血氧饱和度.取心室肌按Nitro-BT法测琥珀酸脱氢酶,以硝酸铅法Ca~(2 )-Mg~(2 )-腺苷三磷酸酶,并通过电镜观察心肌超微结构变化.结果表明,当吸入氧浓度等于或小于8%时,琥珀酸脱氢酶损伤反应明显;等于或小于12%时.Ca~(2 )-Mg~(2 )-腺苷三磷酸酶活性明显降低;当吸入氧浓度为4%时,心肌超微结构出现明显地不可逆性改变.结果提示,当小鼠吸入等于或小于12%的氧15min时,心肌出现明显的代谢障碍.

Abstract:
40 adult mice, both male and female, were divided into 4 groups and confined in a sealed container in which normal oxygen concentration, 12%. 8% and 4% oxygen concentration were supplied for 10-15 min. The partial pressure of oxygen (PaO2) and oxygen satruration (SaO2) in the artery blood were measured by collecting blood from the heart. The activity of succinic de-hydrogenase (SDH) in ventricle myocardium was measured with Nitro-BT method, and Ca2--Mg2--ATPase with lead nitrate method. The change in the ultrastructure of the myocardium was also observed with electron microscopy. The results indicated: When the inhaled oxygen concentration was equal to or less than 8%, the injury in SDH was obvious; When the concentration was equal to or less than 12%, the activity of Ca2 -Mg2 -ATPase was obviously decreased; When the concentration droped to 4%, an irreversible change in the myocardium ultrastructure was obviously observed. The data suggest that if mice are forced to inhale air of the oxygen concentration equal to or less than 12% for 15 min, the disorder of metabolism in the myocardium will occur.
