一氧化氮在血管内皮生长因子效应中作用的研究

摘    要:目的 观察血管内皮生长因子 (VEGF)对兔离体主动脉内细胞增殖的影响 ,了解一氧化氮 (NO)在其中所起的作用 ,明确 VEGF的作用机制。方法 先进行兔主动脉内皮细胞的培养和鉴定 ,然后行传代培养。在第 3~ 5代主动脉内皮细胞培养液中加入不同浓度的 VEGF培养 4 8h,测定主动脉内皮细胞的吸收光密度 (OD)值来观察其增殖程度。并应用一氧化氮合成酶 (NOS)抑制剂 N-硝基 - L -精氨酸甲酯 (L - NAME)后观察内皮细胞 OD值的变化情况。结果 VEGF可使血管内皮细胞 (VEC)的 OD值的升高 ,并呈剂量依赖性 (r=0 .972 7;P<0 .0 1) ,L - NAME可剂量依赖性地抑制其效应 (P<0 .0 1)。结论  NO在 VEGF促 VEC增殖中起中介作用。

Abstract:Objective To observe the biological effect of vascular endothelial growth factor(VEGF) on proliferation of vascular endothelial cells of rabbit aorta inVitro,and to elucidate the role of nitrogen monoxide in the process and the mechanism of VEGF.Methods The vascular endothelial cells of rabbit aorta were cultured and identified,then passage culture of the cell was carried out.The third and the fifth generations of the vascular endothelial cells were cultuered for 48 hours in culture medium with different concentrations of VEGF and proliferation grades of the cells were evaluated by absorptive optical density(OD), then N nitro L arginine methyl ester(LNAME),inhibitory agent of nitrogen monoxide synthetase,was added into the medium and proliferation grades were evaluated as before.Results VEGF could increase the OD value of the vascular endothelial cells dose-dependently(r=0.9727;P<0.01),while LNAME could inhibit the VEGF's effect dose dependently(P<0.01).Conclusion Nitrogen monoxide could play an important role in the process of stimulating proliferation of vascular endothelial cells by VEGF.
