食管鳞状细胞癌中PCNA及突变型P53蛋白的表达及其意义

目的探讨食管鳞状细胞癌中PCNA及突变型P53蛋白的表达及其意义。方法应用流式细胞术（FCM）检测60例食管鳞状细胞癌及食管正常组织中PCNA及突变型P53蛋白的表达水平,并检测食管癌组织中PCNA及突变型P53蛋白表达与不同临床参数之间的关系。分析食管癌组织中PCNA与突变型P53蛋白表达的相关性。FCM方法检测60例食管鳞状细胞癌及食管正常组织细胞增殖情况。结果流式细胞术方法检测到食管癌组织中PCNA与突变型P53蛋白表达水平显著高于正常食管组织（P〈0.05）。食管癌组织中PCNA与突变型P53蛋白表达水平与食管鳞癌细胞分化程度、有无淋巴结转移、癌浸润深度有关（P〈0.05）,与患者的性别及年龄无关（P〉0.05）。食管癌组织中PCNA与突变型P53蛋白表达呈正相关性。结论 PCNA与突变型P53蛋白在食管癌组织中的高表达可能参与了食管癌的发生与发展,PCNA与突变型P53蛋白高表达的食管癌恶性度高,联合检测PCNA与突变型P53蛋白可以为食管癌诊断及判断恶性度提供临床依据。

Expression and significance of PCNA and mutant type P53 protein in patients with esophageal squamous cell carcinoma

Objective To investigate the expression and significance of PCNA and mutant type P53 protein in patients with esophageal squamous cell carcinoma,and to explore initially the correlation between the expression of PCNA,mutant type P53 protein and the pathogenesis and development of esophageal squamous cell carcinoma,and the correlation between the expression of PCNA and mutant type P53 protein in the tissues of esophageal squamous cell carcinoma.Methods The expression levels of PCNA,mutant type P53 protein and cell cycle in 60 cases of normal esophageal tissues and esophageal carcinoma tissues were detected by flow cytometry.The relationship between PCNA,mutant type P53 protein expression and clinical pathological characteristics of esophageal squamous cell carcinoma was analyzed by flow cytometry.The relationship between PCNA and mutant type P53 protein expression in esophageal cancer was analyzed by SPSS.Results The expression levels of PCNA,mutant type P53 protein in esophageal squamous cell carcinoma were significantly higher than those in normal esophageal tissues（P0.05）.The expression levels of PCNA and mutant type P53 protein were closely correlated with the degrees of differentiation,metastasis of lymph node and the invasive depth of esophageal squamous cell carcinoma（P0.05）,however,which were not correlated with patients＇ age and gender（P0.05）.The expression of PCNA was positively related with the expression of mutant type P53 protein in esophageal cancer tissue.Conclusion The overexpression of PCNA and mutant type P53 may be involved in the carcinogenesis and progression of esophageal squamous cell carcinoma.The esophageal cancer with overexpression of PCNA and mutant type P53 showes high malignant degree.Combined detection of PCNA with mutant type P53 protein can provide clinical evidence for the diagnosis of esophageal cancer and judgment of malignant degrees.
