小檗碱联合卡托普利预处理对心肌缺血再灌注损伤的保护作用

摘    要:目的研究小檗碱联合卡托普利预处理对大鼠心肌缺血再灌注损伤的保护作用及其相关机制。方法 SD大鼠随机分为5组,每组6只:假手术组,模型组,小檗碱组,卡托普利组,联合用药组。用BL-420生物采集系统记录各组大鼠心率(HR)、左心室收缩压(LVSP)、左心室舒张末压(LVDEP)、左心室压力变化速率(±dp/dtm ax);用试剂盒测量血清中肌酸激酶同工酶(CK-MB)和乳酸脱氢酶(LDH)活性;以及心肌组织中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性。结果联合用药组可明显降低缺血再灌注大鼠血清中CK-MB和LDH活性(P〈0.01),降低缺血再灌注大鼠心肌组织MDA含量(P〈0.05),提高心肌组织SOD活性(P〈0.05),联合应用组上述作用均比单独用药时增强(P〈0.01)。结论小檗碱联合卡托普利预处理可增强对大鼠缺血再灌注损伤的抗氧化保护作用。

The protective effect of berberine combined with captopril preconditioning on myocardium ischemia/reperfusion injury in rats

Abstract:Objective To investigate the protective effect of berberine combined with captopril preconditioning on myocardium ischemia/reperfusion injury in rats,and to explore its action mechanism.Methods The SD rats were randomly divided into five groups: sham-operation group,model group,berberine group,captopril group;drug combination group.The levels of LVSP,LVEDP and ±dp/dtmax were recorded by BL-420 system.The levels of LDH and CK-MB in serum were detected by using kits.The activity of SOD and the content of MDA in myocardial tissues were detected by using kits.Results Berberine combined with captopril increased the levels of LVSP,±dp/dtmax,however,which decreased the levels of LVEDP.The combination appilication of berberine with captopril decreased the activities of LDH and CK-MB in serum,the content of MDA in myocardial tissues,however,which increased the activity of SOD in myocardial tissues.The effect of berberine combined with captopril was superior to that of berberine or captopril alone.Conclusion The berberine combined with captopril preconditioning can increase the anti-oxidant protective effect on myocardium ischemia/reperfusion injury in rats.
