小檗碱对L-甲状腺素诱发心肌肥厚大鼠心血管功能的影响

摘    要:目的研究小檗碱对皮下注射L-甲状腺素所致心肌肥厚模型大鼠心血管功能的影响。方法SD大鼠皮下注射L-甲状腺素制备心肌肥厚模型,小檗碱分别按5、10mg/kg灌胃给药,连续2周。期间监测动物血压;给药2周后,经左心室插管测量心功能;取心肌组织和血清测定NO含量。结果小檗碱5、10mg/kg均可显著降低L-甲状腺素模型大鼠的血压;小檗碱10mg/kg可减慢心率(HR),降低左心室最大收缩压(LVSP),升高左心室舒张末期压力(LVEDP),降低最大收缩速率( dp/dtmax);并增加左心室肌组织及血清NO含量;且左心室肌组织NO含量与LVSP、LVEDP、 dp/dtmax、HR呈显著的相关性。结论小檗碱对L-甲状腺素诱导的心肌肥厚模型大鼠的血压升高及心功能障碍有保护作用,其作用与增加NO含量具有显著的相关性。

Abstract:Objective To study the influence of berberine (Ber) on cardiovascular function in rats with hypertrophic cardiomyopathy (HCM) induced by L-thyroxin.Methods The model with HCM was established by hypodermic injection of L-thyroxin in SD rats.Ber was given to rats orally (5 or 10 mg/kg) for two weeks, and rat blood pressure was measured.Two weeks later the rat cardiac function was examined by catheterization in left ventricle of heart and nitrous oxide (NO) contents in serum and myocardium were determined.Result Ber (5 or 10 mg/kg)decreased the blood pressure of rat model remarkably and Ber (10 mg/kg) can reduce heart rate (HR), the maximum left ventricle systolic pressure (LVSP), increased left ventricle end-diastolic pressure (LVEDP), reduced dp/dtmax, and increased NO content of left ventricle muscle. The NO content of left ventricle muscle was correlated to LVSP, LCEDP, dp/dtmax, and HR.Conclusion Ber can prevent rats of HCM model from blood pressure increase and heart disorder induced by L-thyroxin. This effect is correlated to the increase of NO content.
