摘    要:目的研究木犀草素对人表皮共培养体系(黑素细胞与角质形成细胞)酪氨酸酶活性及黑素生成的影响。方法培养人表皮黑素细胞和人表皮黑素细胞与角质形成细胞共培养体系,利用四甲基偶氮唑蓝(MTT)还原法测定细胞活力,490nm比色法测定酪氨酸酶活性,475nm比色法测定黑素含量。结果 0.5μmol/L、0.75μmol/L和1μmol/L木犀草素对于黑素细胞及共培养细胞酪氨酸酶活性及黑素生成均有较强的剂量相关性增强作用,0.75μmol/L及1μmol/L木犀草素与对照组相比差异有统计学意义(P〈0.01)。结论木犀草素对于共培养体系酪氨酸酶活性及黑素生成有较强的剂量相关性增强作用。

Effects of Luteolin on the Tyrosinase Activity and Melanogenesis in Co-culture Model of Human melanocytes and keratinocytes

Abstract:Objective To study the effects of luteolin on the tyrosinase activity and melanogenesis in co-culture model of human melanocytes and keratinocytes.Methods The model of co-culture of human melanocytes and keratinocytes was cultured and MTT was used to measure the proliferation of melanocytes and the co-cultures.The tyrosinase activity and melanin content were measured.Results The melanin synthesis and tyrosinase activity were clearly enhanced by luteolin in a dose-dependent manner at concentrations of 0.5μM,0.75μM,and 1μM in both melanocytes and co-cultures.0.75μmol/L and 1μmol/L of luteolin showed statistically significant suppression(P0.01)than the control.Conclusions Luteolin exhibits a a dose-dependent enhancing effects on melanin synthesis and tyrosinase activity in co-cultures of human melanocytes and keratinocytes.
