电针对脊髓损伤后一氧化氮合酶表达的影响

摘    要:目的探讨电针对脊髓损伤后一氧化氮合酶(nitricoxidesynthase,NOS)基因及其蛋白表达的影响。方法采用改良的Allen's垂击法致大鼠脊髓(T10)中度损伤,损伤后即刻进行督脉电针治疗,于术后1、4、24、48h处死动物,分别用RT-PCR法和NADPH-黄递酶组织化学染色法测定各型NOSmRNA及其蛋白的表达,并进行图像定量统计学分析。结果电针可不同程度地降低nNOSmRNA、iNOSm-RNA的表达,降低NOS活性。结论电针早期治疗可能通过降低一氧化氮(NO)的形成减轻脊髓继发性损伤,有一定的神经保护作用。

Abstract:Objective To study the effect of electric-acupuncture on nitric oxide synthase gene and protein expression in rat spinal cord injury. Method The rats were treated in electric-acupuncture after spinal cord injury(T10) by Allen's weight-drop (50 g-cm force) technique and killed at 1, 4, 24, 48 h after operation. Each NOSmRNA and protein of NOS were assayed by RT-PCR, NADPH-Diaphorase histochemistry and image analysis system. Result The expression of nNOSmRNA, iNOSmRNA and activity of NOS were downregulated by electric acupuncture. Conclusion Electric-acupuncture may prevent the progress of the secondary spinal cord injury by decreasing the synthesis of NO and have some protection role on neuronal function.
