丹参与甲基泼尼松龙联用治疗急性肺损伤的实验研究

摘    要:目的探讨丹参和甲基泼尼松龙联合应用对内毒素(lipopolysaccharide,LPS)所致急性肺损伤(acute lung injury,ALI)大鼠的治疗作用及其机制。方法建立大鼠LPS吸人性ALI模型(LPS3mg/kg气管内注射)。50只大鼠随机分为5组:生理盐水对照组;LPS损伤组;甲基泼尼松龙组(LPS+甲基泼尼松龙);丹参组(LPS+丹参);联合用药组(LPS+甲基泼尼松龙+丹参)。观察支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中蛋白含量、肺脏的大体、光镜下病理改变,以及肺组织湿/干重(W/D)比值,并测定肺组织中一氧化氮(nitric oxide,NO)、丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)和谷胱甘肽过氧化物酶(glutathione peroxidase,GSH—Px)的变化。结果经气道吸入LPS可致大鼠发生ALI。与LPS损伤组相比,单用甲基泼尼松龙或丹参组BALF中蛋白含量、肺组织W/D比值和MDA、NO水平均明显降低(P〈0.05),肺组织SOD、GSH—Px活性则明显升高(P〈0.05),以联合用药组更为明显(P〈0.01)。结论联合应用甲基泼尼松龙和丹参通过抗氧化作用、调整氧化/抗氧化平衡,对ALI大鼠具有较好的治疗作用。

Therapeutic Effects of Combined Radix Salviae Miltiorrhizae Injection and Methylprednisolone on Acute Lung Injury of Rats

Abstract:Objective To observe the effect and mechanism of combined radix salviae rniltiorrhizae (RSM) injection and methylprednisolone on acute lung injury (ALI) induced by lipopolysaccharide (LPS) in rats. Methods ALI models were reproduced by intratracheal instillation of LPS (3 mg/kg). Four hours after LPS instillation, rats were randomly divided into five groups: normal saline group, LPS group, methylprednisolone group, RSM group (LPS + RSM) and combination group (LPS + RSM + methylprednisolone). The contents of protein in bronchoalveolar lavage fluid (BALF) and the general and pathological changes of lungs were observed. The wet and dry weight ratio (W/D) of lung was calculated. The concentrations of nitric oxide (NO) and malondialdehyde (MDA), the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in lung tissues were measured by colorimetry. Results ALI was obviously observed in LPS group. In RSM group, methylprednisolone group and combined drug group, the contents of protein in BALF, the MDA, NO contents in lung tissues, and W/D of lung decreased significantly (P〈0. 05 or P〈0. 01), and the activities of. SOD and GSH-Px increased significantly (P〈0. 05 or P〈0. 01) compared with those of LPS group. Conclusion Combination of RSM injection and methylprednisolone has therapeutic effects on ALI in rats by anti-oxidation and adjusting oxidation/anti-oxidation balance.
