比较两种树突状细胞肿瘤疫苗对小鼠乳腺癌的治疗作用

目的比较热休克法与冻融法制备的树突状细胞肿瘤疫苗对小鼠乳腺癌的治疗效果。方法使用BALB/c小鼠建立EMT6乳腺癌肿瘤动物模型,然后将小鼠随机分为三组,分别注射生理盐水、冻融法树突状细胞肿瘤疫苗、热休克法树突状细胞肿瘤疫苗。定期测量肿瘤的大小,至第21天处死小鼠并称量肿瘤重量。结果注射热休克法树突状细胞肿瘤疫苗组肿瘤生长得到明显抑制,与空白对照组比较P〈0.01,与冻融法树突状细胞肿瘤疫苗组比较P〈0.01。结论热休克法制备的树突状细胞肿瘤疫苗较冻融法制备的树突状细胞肿瘤疫苗对小鼠乳腺癌有更好的治疗效果。

The comparision of therapeutic efficacy of dentritic cell vaccines made by two different ways to murine breast cancer

Objective：To compare the therapeutic efficacy of dentritic cell loaded with tumor peptide made by two different ways： heat-treating way and freeze-thrawing way.Methods： Three groups of mice with EMT6 breast cancer were treated with saline,dendritic cell（DC） vaccine made by heat-treating way and DC vaccine made by freeze-thrawing way respectively.The weight and volume of tumor was measured 21 days later.Results： The tumor size in the DC vaccine made by heat-treating way group was significantly smaller compared with control group（P0.001） and the DC vaccine made by freeze-thrawing way group（P0.001）.Conclusion： The immunotherapy based on DCs loaded with heat-treating peptide is more efficient for murine breast cancer and it can be a promising biological therapy.
