负载As2O3壳聚糖纳米粒对大鼠免疫功能的影响

摘    要:
目的:观察负载三氧化二砷(arsenic trioxide, As2O3)壳聚糖纳米粒(NPCS-As2O3)对大鼠免疫功能的影响,为临床用药提供实验和理论依据。方法将40只SD大鼠随机分为4组:壳聚糖纳米粒组( NPCS组),负载As2 O3壳聚糖纳米粒组( NPCS-As2 O3组),As2 O3组和生理盐水( NS)组。尾静脉注射给药,连用7天后处死大鼠,取脾称重计算脾脏指数并分离脾细胞进行特异性细胞毒T淋巴细胞( CTL)功能检测。结果 As2 O3组CTL活性降低,与NS组相比,差异有统计学意义(P<0.05);NPCS组和NPCS-As2O3组均显示出较高的CTL活性,并且脾脏指数增加,与As2O3组和NS组比较差异均有统计学意义(P<0.05)。结论 As2O3可以降低大鼠脾免疫功能;NPCS可以明显提高大鼠脾免疫功能;NPCS-As2 O3可以减轻As2 O3造成的脾免疫功能损伤,保护大鼠脾的免疫功能。

Abstract:
Objective To explore the effects of As 2 O3 loaded chitosan nanoparticle ( NPCS-As2 O3 ) on the im-mune function in rats , which can provide experimental and theoretical evidence for better clinical therapy .Methods Forty SD rats were randomly divided into four groups:a chitosan nanoparticle (NPCS) group, an As2O3 loaded chitosan nanoparticle ( NPCS-As2 O3 ) group, an As2 O3 group and a normal saline( NS) group.The agents were given via tail vein injection for seven days .The spleens were removed to dertermine spleen index , while spleen cells were separated to test the function of specific cytotoxic T lymphocytes (CTLs).Results The activity of CTL in the As2O3 group was re-markably reduced when compared with that in the control group (P〈0.05).Both tthe NPCS group and the NPCS -As2 O3 group presented stronger activities of CTL and higher values of spleen index than those in the As 2 O3 group and NS group (P〈0.05).Conclusions The immune function in rats can be damaged by As 2O3 but remarkably enhanced by NPCS.NPCS-As2 O3 could relieve the immune damage caused by As 2 O3 .The presence of NPCS -As2 O3 suggests a fea-sible idea of reducing the toxic and side -effects of As2 O3 on clinical tumor therapy .
