山茱萸多糖对胃癌细胞增殖和凋亡的机制

摘    要:目的探讨山茱萸多糖对胃癌AGS细胞增殖和凋亡的影响及分子机制。方法用不同剂量山茱萸多糖灌胃接种AGS细胞的裸鼠,检测裸鼠体内肿瘤体积;TUNEL法检测细胞凋亡指数;胃癌细胞AGS分为对照组、山茱萸多糖低、中、高剂量组、si-NC组、si-ELF3-AS1组、山茱萸多糖+pcDNA组、山茱萸多糖+pcDNA-ELF3-AS1组。MTT检测细胞增殖抑制率;流式细胞术检测细胞凋亡;RT-qPCR检测lncRNA ELF3-AS1表达水平。结果山茱萸多糖处理的裸鼠体内移植瘤体积减少,细胞凋亡指数升高(P<0.05)。山茱萸多糖处理胃癌细胞AGS后,细胞增殖抑制率升高,凋亡率升高,ELF3-AS1表达水平降低,且呈剂量依赖性(P<0.05)。胃癌细胞AGS中ELF3-AS1表达水平升高(P<0.05)。抑制ELF3-AS1表达后,细胞增殖抑制率升高,凋亡率升高(P<0.05)。ELF3-AS1过表达逆转了山茱萸多糖对胃癌AGS细胞增殖和凋亡的作用。结论山茱萸多糖可能通过下调ELF3-AS1抑制胃癌AGS细胞增殖,促进AGS细胞凋亡。

Abstract:Aim To investigate the effect of cornus officinalis polysaccharidesson the proliferation and apoptosis of gastric cancer AGS cell line and its molecular mechanism.Methods Different doses of cornus officinalis polysaccharidess were intragastrically inoculated to nude mice with AGS cells,and the tumor volume in nude mice was detected;TUNEL method was used to detect the apoptosis index in tumor tissues of nude mice.Gastric cancer cells AGS were divided into control group,cornus officinalis polysaccharidess low,medium,and high dose group,si-NC group,si-ELF3-AS1 group,cornus officinalis polysaccharidess+pcDNA group,cornus officinalis polysaccharidess+pcDNA-ELF3-AS1 group.MTT was employed to detect cell proliferation inhibition rate,flow cytometry to detect cell apoptosis,and RT-qPCR to detect lncRNA ELF3-AS1 expression.Results The volume of transplanted tumors in nude mice treated with Fructus Corni polysaccharides decreased,and the apoptotic index increased(P<0.05).After cornus officinalis polysaccharidess treatment of gastric cancer cell AGS,the cell proliferation inhibition rateincreased,the cell apoptotic rate increased,and the expression of ELF3-AS1 decreased,dose-dependently(P<0.05).The expression of ELF3-AS1 in gastric cancer cells AGS increased(P<0.05).After inhibiting the expression of ELF3-AS1,the cell inhibitory rate increased,the apoptotic rate increased(P<0.05).The overexpression of ELF3-AS1 reversed the effects of cornus officinalis polysaccharides on the proliferation and apoptosis of gastric cancer AGS cells.Conclusions Cornus officinalis polysaccharides may inhibit the proliferation of gastric cancer AGS cells and promote AGS cell apoptosis by down-regulating ELF3-AS1.
