c—erbB—2过表达及点突变与喉癌发生的关系

目的：探讨喉癌基因ｃ－ｅｒｂＢ－２点空及过表达同喉癌发生发展的关系。方法：应用ＰＣＲ－ＳＳＣＰ对ｃ－ｅｒｂＢ－２的点突变进行检测，采用免疫组化法分析ｃ－ｅｒｂＢ－２的过表达。结果：４０例喉癌阳性表达率为５２．５％（２１／４０）。其中２４例声门上癌表达率为５７％（１７／３０），６例声带癌表达率为５０（３／６），４例声门下癌表达率为２２．５％（１／４），同前者差异有高度显著性（Ｐ〈０．０１），本见ｃ－

Overexpression and point mutation of c-erbB-2 in laryngeal squamous carcinomas and its carcinogenesis

Objective: To investigate the overexpression and point mutation of c erbB 2 involved in the carcinogenesis process of laryngeal squamous carcinomas.Methods: Using laryngeal squamous carcinoma specimens from 40 patients.The point mutation of c erbB 2 was determined by PCR-SSCP;the over expression of c erbB 2 was analyzed by immunohistochemical methods.Results:Of 40 laryngeal carcinoma specimens,the overexpression of c erbB 2 were detected in 18 cases,the positive staining was 52.5%(21/40);the positive staining of 21 supraglottic carcinomas and of 6 glottic carcinoma were 57% (17/30) and 50%(3/6) respectively,while the positive staining of 4 subglottic carcinoma was only 22.5%(1/4),the difference was significant (P<0.01).In comparison with the former,no point mutation was observed in these specimens.Conclusion:There are extensive overexpression and amplification of c erbB 2 in laryngeal carcinoma,but no detection of point mutation.This suggests that other kind of activation probably has not been found,or the over expression and amplification of c erbB 2 might be the main reason that promotes laryngeal carcinoma.
