DNA甲基化转移酶1在非小细胞肺癌中的表达及临床意义

目的观察DNA甲基化转移酶1(DNMT1)在非小细胞肺癌(NSCLC)组织、癌旁组织及正常肺组织中的表达,通过其表达水平的分析,探讨其与临床特征的关系。方法选取睢宁县人民医院胸外科手术切除的肺癌标本,分别取癌组织54例,癌旁组织48例,另取周边正常组织24例为对照组,行免疫组化检测,分析各组织中DNMT1表达差异及与临床各指标之间关系。结果 DNMT1在NSCLC的癌组织、癌旁组织、正常肺组织中阳性表达率分别39/54(72.2%)、30/48(62.5%)、0/24(0%),与正常肺组织比较差异有统计学意义(P<0.01)。DNMT1的表达与NSCLC组织的分化程度、TNM分期、患者的年龄、性别、吸烟、病理类型无相关性。结论 DNMT1在NSCLC癌组织、癌旁组织中出现高表达,在正常肺组织中表达缺失,提示DNMT1参与NSCLC发病机制,可以作为病理诊断NSCLC的标志物。DNMT1与NSCLC的临床特征之间无相关性,不能作为预后的指标。

The expression and clinical significance of DNA methylation transferase enzyme 1 in non-small cell lung cancer

Objective To investigate the expression of DNMT1(DNA methylation-transferase enzyme 1(DNMT1) in human non-small cell lung cancer(NSCLC) and analyze the relationship with the clinical characteristic.Methods Fifty-four samples were obtained from patients with NSCLC and had surgery in Suining People′s Hospital.Immunohistochemistry(sABC) was used to detect the expression of DNMT1 protein in 54 cancerous,48 peficancerous tissues and 24 adjuvant normal tissues.Results The positive rates of DNMT1 of NSCLC and peficancerous group were significantly higher than those of peripheral normal lung tissues.The positive rates were 39/54(72.2%),30/48(62.5%),0/24(0%) respectively.Compared with normal lung tissue,the difference was statistically significant(P<0.01).DNMT1 expression had no significant relationship with cell differentiation,sex,age,smoking history,pathologic type of NSCLC patients.Conclusion The higher expression of the DNMT1 in NSCLC suggests the protein participate in carcinogenesis and can be used as characteristic parameters for the diagnosis of NSCLC.There is no statistically significant correlation between DNMT1 expression and age,sex,smoking history,pathologic type.The protein can not be used as characteristic parameters for the prognosis of NSCLC.
