摘    要:目的:研究红细胞膜蛋白的变化对膜脂流动性的影响。方法:从三个不同方面改变膜蛋白结构,即(1)用戊二醛交联膜蛋白分子;(2)用伴刀豆蛋白(ConA)作为配体与红细胞膜上的受体蛋白相作用;(3)用胰蛋白酶降解膜表面蛋白,然后利用荧光分光光度计测量经上述三种方式处理后的红细胞膜的荧光偏振度(P)。结果:经三种方法处理地的红细胞膜的荧光偏振度P值均有所上升,表明膜脂流动性下降。结论:膜蛋白分子的改变将会影响膜脂的流动性。

Abstract:Objective:To investigate the effect of change of RBC membrane proteins in structure on the fluidity of the membrane lipid.Methods:By using three kinds of methods to change RBC membrane proteins.which are follows:(1)making the RBC membrane proteins linked with glutaraldehyde.(2)causing the interactions between the RBC membrane receptors and ligands with concanavalin A(ConA).(3)degrading the RBC membrane proteins with trypsin.Then we measured the lipid fluidity of the RBC membrane treated with the above ways by fluorescence polarization technique.Results:The lipid fluidity declined distinctly for the RBC membrane which were treated by the three kinds of methods above.Conclusion:The decrease of membrane lipid fluidity may results from the conformation changes of RBC membrane proteins.
