反义IGF—I基因转染抗小鼠B16黑色素瘤的研究

摘    要:
利用基因治疗方法中反义RNA技术,通过脂质体包裹反义胰岛素生长因子-I(IGF-I)基因导入小鼠B16黑色素瘤,与对照组相比致瘤性降低,生长缓慢,荷瘤鼠的胸腺重量增大,脾脏的生发中心细胞增殖,白髓和红髓的淋巴细胞扩增的程度很明显,体内反义IGF-I基因治疗与LAK细胞治疗联合应用延长荷瘤小鼠的生存期的效果胜过两者单独应用的效果,结果表明:反义IGF-I基因具有一定的抑制小鼠B16黑色素生长的作用和

Abstract:
By applying the antisense strategy of gene therapy,antisense insulin like growth factor I(IGF I) gene was transfected into mouse B16 melanoma by the method of Lipofectin mediated gene transfer The tumorgenic phenotype of mice with transfectants was decreased in contrast with nontransfectants The thymus weight of mice with transfectants became larger The germinal centers appeared in spleen tisseue The lymphocytes proliferation of white pulp and red pulp was obvious The gene therapy of antisense IGF I for mouse B16 melanoma had de finite curative effect,moreover,in combination with LAK cell treatment,the curative effect was more marked The results showed the application of antisense IGF I gene could inhibit the growth of B16 melanoma and enhance tumor cell immunogenicity,it provided a new approach to the gene therapy in clinical practice
