口腔鳞癌中染色体9q22-31的杂合性丢失

摘    要:目的 检测染色体9q在口腔鳞癌发生发展中的作用。方法 应用聚合酶链反应检测22例经显微切割的口腔鳞癌组织中DNA微卫星多态标记D9S299(9q22-31)。结果 杂合性丢失检出率为43.8%(7/16),其中高分化鳞癌为33.3%,而低分化鳞癌为75%。结论 染色体9q22-31区域在口腔鳞癌发生发展中起重要作用。提示此区域可能存在与口腔鳞癌发生发展密切相关的抑癌基因。

Abstract:Objective To investigate the potential loss of tumor suppressor gene loci on chromosome 9q in oral squamous cell carcinogenesis. Methods 22 paired samples of normal and squamous cell carcinoma from the same patients were analyzed. Tissue microdissection under direct microscopic visualization procured pure populations of squamous cell. LOH was detected by examining microsatellite polymorphic sequences in loci D9S299(9q22-31) by using PCR. Results In 7 of 16(43.8%), LOH was found at 9q22-31(D9S299). The frequency of LOH in the well-differentiated type is 33.3% , while in the poorly differentiated is 75%. Conclusion It suggested that the deletion of chromosome 9q22-31 is probably related to the oral squamous cell carcinogenesis.
