慢性粒细胞白血病患者骨髓中不同细胞诱生树突状细胞的体外研究

摘    要:目的:体外诱导慢性粒细胞白血病(CML)患者骨髓单个核细胞(BMMNCs)、CD34+细胞、单核细胞为树突状细胞(DCs),并对其形态、表型及其刺激T细胞增殖作用进行比较。方法:利用GM-CSF、IL-4、TNF-α体外定向诱导生成树突状细胞,利用流式细胞仪对所诱生的细胞进行细胞形态及表型检测,用D-FISH方法检测所诱生DCs的白血病源性。并应用MTT法检测所诱生的DCs刺激T细胞增殖的能力。结果:CML患者BMMNCs、CD34+细胞、单核细胞体外均可诱导生成bcr/abl融合基因阳性的DCs,前者DCs产量明显高于后二者,但均具有刺激自体及异体T细胞增殖的能力,且三者无显著性差异。结论:来源于CML患者骨髓的BMMNCs、CD34+细胞、单核细胞均可诱生成CMLDCs,对T细胞均具有刺激增殖能力,但BMMNCs所诱生的DC主要来源于未成熟的中性粒细胞。

Abstract:Objective To induced dendritic cell(DCs)from bone marrow mononuclear cells (BMMNC),CD34 + hematopoietic cells,monocytes of patients with chronic leukemia(CML).To compare the differences between them.Method The cells were induced by GM-CSF,IL-4,TNF-αin vitro for 14days.The induced cells were detected for morphological and phenotypic properties by in-versed microscope,electro-microscope and FACS.The induced cells were detected for bcr /abl fusion gene by D-FISH.The ability of induced cells to stimulate proliferation of T lymphocyte was tested by MTT test.Results BMMNCs,CD34 + hematopoietic cells,mono-cytes of patients with CML could be induced by IL-4,GM-CSF,TNF-αinto DCs with bcr /abl fusion gene that observed.DCs de-rived from the three kinds of cells all had the ability to stimulate proliferation of autogenic or allogenic T cells.The number of DCs derived from BMMNCs was much more than the others.Conclusion DCs derived from BMMNCs of patients with CML mainly were derived from immatural neutrophils.
