广藿香抑制肠推进药效指纹图谱构建与谱效关系研究

摘    要:目的 构建广藿香抑制肠推进的高效液相色谱(HPLC)药效指纹图谱,并分析图谱中各成分与抑制肠推进作用的相关性.方法 以乙酸乙酯、丙酮、甲醇、水分别提取广藿香,提取物分别行小鼠肠推进实验,筛选药效最佳者,以HPLC法建立指纹图谱,以夹角余弦法计算相似度,以灰色关联法分析图谱中各特征峰峰面积与肠推进效应的关联度大小.结果 广藿香甲醇提取物对小鼠胃肠蠕动抑制作用最强,此基础上建立10批广藿香药材药效指纹图谱,相似度均大于0.81,共有18个特征峰,特征峰P5、P10、P12、P13和P14对应成分与药效关联度最大,顺序为P14> P13> P5> P12>P10.结论 广藿香抑制肠推进药效指纹图谱应在甲醇提取物基础上建立,该抑制作用主要由多种成分主导.

Abstract:Objective To establish pharmacodynamic fingerprint of Herba Pogostemonis with HPLC and study its correlation with anti-gastrointestinal propulsive motility.Methods Herba Pogostemonis was extracted respectively by ethyl acetate,acetone,methanol and water.Extracts were tested by gastrointestinal propulsive experiment,HPLC fingerprint was set up based on the extract of best pharmacological effects.The spectrum-effect relationship was studied by grey relation statistic.Results Methanol extract showed strongest inhibition of gastrointestinal propulsive motility.On this basis,pharmacodynamic fingerprint with 18 characteristic peaks was set up.Corresponding components of P5,P10,P12,P13 and P14 showed maximum correlation with pharmacological effects,the size order was P14 〉 P13 〉 P5 〉 P12 〉 P10.Conclusion The pharmacodynamic fingerprint of Herba Pogostemonis should set up based on the methanol extract.The anti-gastrointestinal propulsive effect of Herba Pogostemonis is related to a variety of ingredients.
