HPLC法测定抗心衰颗粒中黄芪甲苷的含量

目的：用高效液相色谱法测定抗心衰颗粒中黄芪甲苷的含量。方法：以吡啶-苯甲酰氯（2．5：1）为衍生化试剂，采用Supelco ODS C18柱（250mm×4．6mm，5μm），流动相为甲醇-四氢呋喃-0．2％三乙胺水溶液（90：4：6），流速为1．2ml·min^-1，检测波长为230nm。结果：黄芪甲苷衍生物进样量在0．03～0．93μg范围内与峰面积积分值呈良好线性关系（r=0．9999），平均加样回收率为95．17％，RSD=2．31％（n=6）。结论：该方法简便易行，结果准确，重复性好，可用于测定抗心衰颗粒中黄芪甲苷的含量。

Determination of Astragaloside IV in Kangxinshuai Granules by HPLC

Objective： To establish a method for the determination of astragaloside IV in Kangxinshuai granules by HPLC. Method： The derivatizing reagent was benzoyl chloride in pyridine （ 1： 2.5, V/V）. The Supelco ODS C18 column （250 mm × 4.6 mm,5 μm） was used with methanol-tetrahydrofuran -0.2% triethylamine water solution（90： 4： 6） as the mobile phase. The flow rate was 1.2 ml· min^- 1, and the detection wavelength was at 230 nm. Result： The linear range of derivatization of astragaloside Ⅳ was 0.03 μg -0.93 μg （ r = 0. 999 9）, the average recovery rate was 95.17% , and RSD was 2.31% （ n = 6）. Conclusion： The method is simple, rapid, accurate and reproducible. It can be used for the content determination of astragaloside Ⅳ in Kangxinshuai Granules.
