摘    要:
目的:用高效液相色谱法测定抗心衰颗粒中黄芪甲苷的含量。方法:以吡啶-苯甲酰氯(2.5:1)为衍生化试剂,采用Supelco ODS C18柱(250mm×4.6mm,5μm),流动相为甲醇-四氢呋喃-0.2%三乙胺水溶液(90:4:6),流速为1.2ml·min^-1,检测波长为230nm。结果:黄芪甲苷衍生物进样量在0.03~0.93μg范围内与峰面积积分值呈良好线性关系(r=0.9999),平均加样回收率为95.17%,RSD=2.31%(n=6)。结论:该方法简便易行,结果准确,重复性好,可用于测定抗心衰颗粒中黄芪甲苷的含量。

Abstract:
Objective: To establish a method for the determination of astragaloside IV in Kangxinshuai granules by HPLC. Method: The derivatizing reagent was benzoyl chloride in pyridine ( 1: 2.5, V/V). The Supelco ODS C18 column (250 mm × 4.6 mm,5 μm) was used with methanol-tetrahydrofuran -0.2% triethylamine water solution(90: 4: 6) as the mobile phase. The flow rate was 1.2 ml· min^- 1, and the detection wavelength was at 230 nm. Result: The linear range of derivatization of astragaloside Ⅳ was 0.03 μg -0.93 μg ( r = 0. 999 9), the average recovery rate was 95.17% , and RSD was 2.31% ( n = 6). Conclusion: The method is simple, rapid, accurate and reproducible. It can be used for the content determination of astragaloside Ⅳ in Kangxinshuai Granules.
