青霉素酰化酶的分离和纯化

摘    要:采用渗透法冲击法提取大肠杆菌E.coli108青霉素酰化酶,得到了高比活的粗酶液,收率为93.3%,经硫酸铵分级沉淀、DEAE-Cellulose-52离子交换柱层析不得到了聚丙烯酰胺凝胶电泳显示一条单带的酶,酶液比活为28.09u/mg,提纯了5.32倍,纯酶总收率为36.5%。

Abstract:Penicillin G acylase from E.coli 108 was extracted by osmotic shock method to obtain crude enzyme of high specific activity with total recovery of enzyme activity of 93.3%. The enzyme was purified by means of (NH 4) 2SO 4 fractional precipitation and ion exchange column chromatography on DEAE Cellulose 52 to give a product with a single band on polyacrylamide gel electrophoresis. The speci fic a ctivity, purification and recovery of enzyme activity were 28.09 u/mg, 5.32 fold and 36.5%, respectively.
