RUNX3、Bcl-2和Bax蛋白在乳腺癌中的表达及其意义

摘    要:目的:检测乳腺癌组织中RUNX3、Bcl-2和Bax基因的表达,探讨它们的相关性及其临床意义。方法:采用免疫组织化学S-P法检测上述基因的表达。结果:RUNX3在乳腺癌中的阳性表达率为35.23%,明显低于在乳腺纤维腺瘤(85%)及乳腺增生病(87.5%)组织中的表达率,差别具有统计学意义(P〈0.05)。Bcl-2和Bax蛋白在良恶性乳腺病组织中的表达差别无统计学意义。RUNX3和Bcl-2蛋白表达与乳腺癌的临床分期、淋巴结转移呈负相关,Bcl-2蛋白表达与病理分级呈负相关。Bax蛋白的表达与临床分期、病理分级及淋巴结转移呈正相关。乳腺癌中RUNX3与Bcl-2蛋白的表达呈正相关(P〈0.05),与Bax的表达无相关性(P〉0.05)。结论:三种蛋白对乳腺癌的发生发展起重要作用,在乳腺癌发展中RUNX3与Bcl-2起协同作用,检测乳腺癌组织中RUNX3、Bcl-2和Bax基因的表达对于评价患者的预后有一定价值。

Abstract:Objective:To study the expression of RUNX3,Bcl-2 and Bax in human breast cancer and its significance.Methods:Immunohistochemical S-P method was used in this study.Results:The positive rate of RUNX3 protein in breast cancer was 35.23%.It was significantly lower than that in breast fibroadenoma(85%) and breast fibrocytic disease(87.5%).There was not a statistically significant difference in the expression of Bcl-2 and Bax protein between the benign and malignant breast tumor.The positive rate of RUNX3 and Bcl-2 was negatively correlated to the clinical stage or node metastasis(P<0.05).There was a positive correlation between the positive rate of Bax and clinical stage,histologic grade or node metastasis(P<0.05).The expression of RUNX3 was positively correlated with the expression of Bcl-2,but not with Bax.Conclusion:RUNX3,Bcl-2 and Bax may play an important role in the carcinogenesis and progression of breast cancer.RUNX3 and Bcl-2 may play synergetic roles in the process of breast cancer.The detection of RUNX3,Bcl-2 and Bax protein has a definite value in judging prognosis in breast cancer.
