摘    要:目的:观察三七总皂苷(PNS)对肝纤维化大鼠的保护作用。方法:SD大鼠随机分成正常组、模型组、PNS(50、100、200mg/kg)组和秋水仙碱(C01,0.1mg/kg)组。除正常组外,其余各组采用皮下注射50%四氯化碳(CCl4),每周2次,连续18周的方法诱导肝纤维化大鼠模型。于造模第9周起,给药组分别灌胃给予相应的受试药物,正常组和模型组给予等量溶媒,每天1次,连续10周。实验结束后,比色法测定血清中超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH)的水平;同时取固定部位肝脏组织,HE染色观察肝纤维化程度;免疫组织化学法检测肝组织胶原I(CollagenI)蛋白的表达,RT—PCR技术测定CollagenImRNA的表达。结果:与模型组相比,PNS(100,200mg/kg)不仅可减轻大鼠肝纤维化程度,降低MDA含量,升高SOD、GSH水平,还可抑制肝纤维化大鼠肝组织中CollagenI的表达。结论:PNS对大鼠肝纤维化具有一定的保护作用。

Abstract:AIM: To observe the effect of Pa-nax notoginseng saponins (PNS) on hepatic fi brosis in rats. METHODS: The rats were ran-domly divided into six groups, including normal group, liver fibrosis model group, PNS (50, 100, 200 mg/kg) treated groups and Col (0.1 mg/kg) group. Rat liver fibrosis was induced by subcutaneous injection 50% carbon tetrachloride (CCI,) twice a week for 18 weeks. PNS were used daily via lavage at 9th week for subsequent 10 weeks. The superoxide dismutase (SOD), malondialdehyde (MDA) and glutathione peroxi dase (GSH) were determinate by chromatome try. Moreover, liver samples were taken to ex amine the degree of liver fibrosis by HE stainingand to stain by immunoehemistry of Collagen I. Collagen I mRNA expression was detected by RT-PCR. RESULTS: As compared with the fi- brotie model group, PNS (100, 200 mg/kg) sig nificantly reduced histopathological change, de- graded MDA contents, raised the levels of SOD and GSH. Meanwhile, PNS could efectively de- crease the expression of Collagen I in liver fibro sis rats. CONCLUSION: PNS has protective effect on hepatic fibrosis in rats.
