摘    要:目的研究大孔吸附树脂分离纯化山楂总黄酮的工艺条件。方法以紫外可见分光光度法测定的山楂样品溶液中总黄酮的含量为指标,采用正交试验和单因素考察相结合的方法考察多个工艺参数。结果D101型大孔树脂对山楂的总黄酮有良好的吸附,其吸附分离工艺条件的药液浓度为0.1g.ml-1、pH3~4,上样量为4倍柱体积,以1m l.min-1吸附速率进行吸附,4.5倍柱体积的80%乙醇洗脱效果最佳。结论方法简单易行,分离效果良好,适于山楂中总黄酮的分离纯化。

Abstract:Objective: To study the technique environment for separating and purifying the total falvones from Crataegus Pinnatifida Bge by Macroreticular Adsorbents.Methods: The total falvones content,in the sample solution of Crataegus Pinnatifida Bge,detected by UV Spectrophotometry,is used as the index.Some technological parameters are observed by the combined methods of orthogonal teat and single factor observation.Results: Macroreticular Adsorbents of D101 type has good absorbing effect on the total falvounes of Crataegus Pinnatifida Bge.The liquid concentration of its absorbing and separating technological environment is 0.1g.ml-1,ph3-4,and the volume of 4 bed volume,and at the absorbing speed of 1ml.min-1.The elution effect of 80% alcohol of 4.5 bed volume is the best.Conclusions: This method is simple and feasible,fit for separating and purifying the total falvones from Cratargus Pinnatifida Bge.
