草乌煎煮时间、给药剂量与毒性的相关性研究

摘    要:
目的:揭示草乌煎煮时间、给药剂量与毒性的相关性.方法:采用均匀设计的方法,将草乌分别煎煮15 min、30 min、1 h、2 h、3 h、4 h、6 h,生药含量依次为临床用量的6倍、12倍、24倍、48倍、72倍、96倍、120倍.模型组和给药组于复制痹证动物模型后,按组给药30d.观测动物生长发育、血液学、血液生化学、脏器系数、病理组织学变化,及停药15 d后上述指标的变化.将结果进行多元回归分析,得出草乌煎煮时间、剂量与毒性的相关性.结果:草乌水煎液具有抗炎镇痛作用;对模型动物的生长发育无明显影响;对其造血系统无明显毒性;对其肝脏、肾脏、心脏有一定毒性.毒性最大的煎煮时间和给药剂量是30 min、120倍,药效最佳的煎煮时间和给药剂量也是30 min、120倍.结论:煎煮30 min,给药剂量为临床用量120倍时,草乌水煎液对痹证动物产生的毒性最大,同时药效最佳.

Abstract:
Objective:Revealing the relativity of decocting time(DT),administration dosage(AD) and the toxicity of Radix Aconiti Kusnezoffii(RAK).Methods: Employing Uniform Designed technique,RA had different DT of 15 min,30 min,1 h,2 h,3 h,4 h and 6 h,according to 6times,12times,24times,48times,72times,96times,120times of clinic AD.All rats except normal control group were made up BiZheng animal model,and administered RAK decocted liquids for 30 days.Surveyed variation of hematology,blood biochemistry,anatomied,histopathology test index and above test index after 15 days.The outcomes of Regression Analysis revealed the relativity between DT,AD and the toxicity of RAK.Results:RAK decocted liquids have drug action of resisting inflammation and reliving pain,not apparently effected the growth and development of the BiZheng model animal,and have not apparent toxicity to its hematopoietic system.They have certain toxicity to liver,kidney,and heart of the model animal.The DT and AD of maximal toxicity of RAK decocted liquids are 30 min and 120 times,and the DT and AD of best medicinal effect are also 30 min and 120 times.Conclusions: RAK decocted liquids with DT 30min and AD 120times have maximal toxicity and best medicinal effect.
