肺癌肿瘤抑制因子1在膀胱尿路上皮癌中的表达

目的探讨肺癌肿瘤抑制因子1（tumor supressor in lung cancer 1,TSLC1）与膀胱尿路上皮癌病理分级的相关性。方法免疫Balb/c雌性小鼠,通过脾细胞和骨髓瘤细胞融合技术制备TSLC1单克隆抗体,并测定效价;同时选取膀胱尿路上皮癌蜡块65例（低危组40例,高危组25例）,正常膀胱黏膜20例,应用免疫组化检测TSLC1蛋白表达情况,由两位病理医生根据染色情况确定阴性（-）、弱阳性（＋）、强阳性（＋＋）。利用SPSS 13.0软件对不同病理分级的TSLC1蛋白表达情况进行统计学分析。结果成功制备TSLC1单克隆抗体IgG1,测其效价为1∶100 000;免疫组化示细胞质、细胞膜显色。高危组TSLC1蛋白表达阴性为88%（22/25）,弱阳性为12%（3/25）;低危组TSLC1蛋白表达弱阳性为75%（30/40）,阴性25%（10/40）;正常膀胱黏膜移行上皮均为强阳性表达,三组间差异均有统计学意义（P〈0.05）。结论TSLC1蛋白的表达与膀胱尿路上皮癌病理分级呈负相关,在评价肿瘤预后方面具有重要的临床参考价值,是膀胱肿瘤生物学行为中有价值的标志物之一。

The expression of TSLC1 in bladder urothelial carcinoma

Objective To study the relationship between tumor supressor in lung cancer 1（TSLC1） and pathological grade of bladder urothelial carcinoma（BUC）. Methods At first,immunize Balb/c female mice to produce monoclonal antibody agaist TSLC1 by spleen cell and myeloma cell fusion technology,and determine its titer;then we choose 65 BUC pathology wax objects,40 of which are low malignant tumors,the others are high malignant tumors.And we also choose 20 normal mucous membrane tissues of urinary bladder.Then we determine the expression of TSLC1 protein with immunohistochemical technology.The pathology doctors help us to analyze the dyeing states.they determine three states：negative（-）、weakly positive（＋）、strongly positive（＋＋）. Results We prepare the monoclonal antibody agaist TSLC1 successfully,IgG1,the titer is 1∶100 000;The expression result of immunohistochemical technology is in cell plasma and membrane.High malignant BUC samples,the weakly positive（＋） is only 12%（3/25）,most others are negative（22/25）;Low malignant BUC samples,the weakly positive is 75%（30/40）,the others are negative（10/40）,what is more,with all normal mucous membrane of urinary bladder,the TSLC1 protein express in all samples,and they are strongly positive（＋＋）,the difference among the three groups are statistically significant（P〈0.05）. Conclusions The expression of TSLC1 protein showed a negative correlation with the pathological grade of BUC,it has important clinical value in prognosis evaluation,it is a valuable marker in the biological behavior of bladder tumor.
