米屈肼预给药对小鼠急性肺损伤的保护作用

摘    要:
目的: 研究米屈肼对脂多糖所致小鼠急性肺损伤的保护作用。 方法: 雄性ICR小鼠72只,随机分为正常对照组、急性肺损伤模型组、米屈肼预处理组。米屈肼组连续6天经腹腔注射米屈肼(100 mg/kg),正常对照组和模型组均以等量生理盐水代替米屈肼。模型组和米屈肼组以脂多糖(4 mg/kg)经气管内滴入,正常对照组经气管内滴入等量生理盐水,6、12、24 h后处死小鼠,收集小鼠肺组织,测定各组肺湿重/干重比值、肺组织匀浆丙二醛(malondialdehyde, MDA)含量、总超氧化物歧化酶(total superoxide dismutase,T-SOD)活力及中性粒细胞髓过氧化物酶(myeloperoxidase,MPO)含量。 结果: 米屈肼组炎症浸润和出血渗出均较模型组减轻,肺湿重/干重比值(W/D)及肺组织中丙二醛、MPO含量均较模型组明显降低(P<0.05),肺组织总SOD活力则明显升高(P<0.05)。 结论: 米屈肼预处理对脂多糖所致急性肺损伤具有保护作用。

Abstract:
Objective: To observe the potential protective effects of mildronate(THP) on the lung tissue in acute lung injury(ALI) in mice.Methods: The experimental objects were randomly divided into normal control,lipopolysaccharides alone,and lipopolysaccharides+THP groups.Mice in lipopolysaccharides +THP group were intra-abdominal preinjection with THP at dosage of 100 mg/kg.ALI models were duplicated by means of intratracheal instillation of lipopolysaccharides.The anti-oxidant ability was analyzed by malondialdehyde(MDA),total superoxide dismutase(T-SOD) activity,and myeloperoxidase(MPO) assays.The structure of lung was examined by light microscope.Results: Compared with lipopolysaccharides alone group,MDA and MPO were decreased significantly(P<0.05) in normal control group and THP+ lipopolysaccharides group,while T-SOD activity was increased.Meanwhile,pretreatment with THP significantly improved the lung pathologic change.Conclusion: THP can protect the lung tissue from damage by lipopolysaccharides.
