抗日本血吸虫循环抗原单克隆抗体杂交瘤细胞株的建立及鉴定

摘    要:为检测日本血吸虫宿主循环抗原提供有价值的McAb,用小鼠骨髓瘤细胞SP2/O株与日本血吸虫循环抗原免疫的BALB/C小鼠脾细胞进行融合,融合率90.20%。经用ELISA和IFA检测,总阳性率34.13%。经多次筛选和克隆化后,获得2株分泌McAb的细胞株:G-A12和H-E9。它们均属IgG2a亚类。Western blot结果表明G-A 12所针对的靶抗原为sjCAg31KD,H-E9为SjAW 31 KD。IFA检测发现H-E9主要定位于成虫表膜;G-A12主要定位于成虫肠道。这2株杂交瘤细胞在体外培养下稳定地分泌特异性抗体迄今已达6个月。

Abstract:In order to supply valuable monoclonal antibody for detecting circulating antigen in schistosomiasis, SP2/0 myeloma and spleen cells of BALB/c mice immunized with SjCAg were fused. The fusion rate was 90.20% An overall positive rate of 34.13% was obtained by ELISA and IFA. 2 monocloned antibody cell lines (G-A12, H-E9) have been established. The immunoglobulin subclass of 2 McAbs was identified to be IgG2a. Results of Western blot showed that target antigen of G-A and H-E 9 was SjCA31KD and SjAw31KD respectively. IFA test showed that H-E 9 was located mainly on the surface membrane of adult worms, and G-A12 was located mainly on the digestive tract. The 2 cell lines have been stably secreting monoclonal antibodies against S. japonicum in tissue culture for 6 months.
